Assembly of Human Neuronal Nicotinic Receptor α5 Subunits with α3, β2, and β4 Subunits

Abstract: Nicotinic acetylcholine receptors formed from combinations of ␣3, ␤2, ␤4, and ␣5 subunits are found in chicken ciliary ganglion neurons and some human neuroblastoma cell lines. We studied the co-expression of various combinations of cloned human ␣3, ␤2, ␤4, and ␣5 subunits in Xenopus oocytes. Expression on the surface membrane was found only for combinations of ␣3␤2, ␣3␤4, ␣3␤2␣5, and ␣3␤4␣5 subunits but not for other combinations of one, two, or three of these subunits. ␣5 subunits assembled inside the oocyte… Show more

“…Incorporation of the a5 subunit in functional recombinant nAChRs Recombinant nAChRs containing a3 and a5 were fully characterized by Lindstrom and co-workers (Gerzanich et al, 1998;Wang et al, 1996;1998;Nelson & Lindstrom, 1999). Our data agree with their observation that a5 does not a ect the EC 50 or Hill slope of the a3b4 dose-response curve to ACh, but increases apparent desensitization to high ACh concentrations (an e ect not shared by b3, Groot-Kormelink et al, 1998;Boorman et al, 2000).…”

“…As previously reported by Wang et al (1996), expressing the a5 subunit together with a3 and b4 had no apparent e ect on the a3b4 ACh dose-response curve (Table 1). Both the EC 50 and the Hill slope of a3b4+a5 receptors were similar to those of a3b4 nAChR.…”

“…A notable developmental difference is that ␣5 expression in CG neurons becomes dependent on retrograde target-derived signals after postganglionic synapse formation. This is an interesting change because ␣5 subunits may be necessary for the formation of high conductance nAChR channels and mature functional properties (Margiotta and Gurantz, 1989;Ramirez-Latorre et al, 1996;Wang et al, 1996;Sivilotti et al 1997;Yu and Role, 1998). Axotomy also decreases perisynaptic ␣7-nAChR total protein levels by fourfold, but with a more rapid time course relative to that of ␣3-nAChRs (Jacob and Berg, 1987).…”
