Assessing the Detection Limit of a Minority Solid-State Form of a Pharmaceutical by 1H Double-Quantum Magic-Angle Spinning Nuclear Magnetic Resonance Spectroscopy

Abstract: The lower detection limit for 2 distinct crystalline phases by H magic-angle spinning (MAS) solid-state nuclear magnetic resonance (NMR) is investigated for a minority amount of cimetidine (anhydrous polymorph A) in a physical mixture with the anhydrous HCl salt of cimetidine. Specifically, 2-dimensionalH double-quantum (DQ) MAS NMR spectra of polymorph A and the anhydrous HCl salt constitute fingerprints for the presence of each of these solid forms. For solid-state NMR data recorded at a H Larmor frequency o… Show more

“…These high frequency 1 H NMR peaks typically arise from hydrogen atoms in amine, ammonium, and carboxylic acid functional groups. 33,37,[40][41][42] The high frequency 1 H NMR signals can be used to observe the 1 H SSNMR spectra of the APIs within formulations. “…High frequency acid or amine peaks from APIs are typically between 8 and 20 ppm 33,37,[40][41][42] and are minimally affected by SSPs targeting the excipient resonances (Figure S2 and S4). Note that most APIs are marketed as free acids, salts, or cocrystals; 59 therefore, APIs will usually possess amine, ammonium, or acid functional groups that give rise to high frequency 1 H chemical shifts.…”

“…Terahertz pulsed spectroscopy has been reported to detect solid forms with a limit of detection of approximately 1 % [21]. Similarly, solid-state nuclear magnetic resonance (ss-NMR) has a detection limit of 1 % [22]. The approach presented here is based on detecting solid form impurities at a single particle level, which is a fundamentally different approach to these bulk methods.…”
