Assessing the Relationship between Lung Density and Function with Oxygen-Enhanced Magnetic Resonance Imaging in a Mouse Model of Emphysema

Abstract: PurposeA magnetic resonance imaging method is presented that allows for the simultaneous assessment of oxygen delivery, oxygen uptake, and parenchymal density. The technique is applied to a mouse model of porcine pancreatic elastase (PPE) induced lung emphysema in order to investigate how structural changes affect lung function.MethodNine-week-old female C57BL6 mice were instilled with saline or PPE at days 0 and 7. At day 19, oxygen delivery, oxygen uptake, and lung density were quantified from T1 and proton-… Show more

“…In addition, these measurements and their comparability among groups provide evidence that sevoflurane does not significantly alter lung mechanics. Although the mean arterial pressure of all the ventilated mice appear to be lower compared to earlier studies [ 18 ], all other parameters analyzed lie within the normal range for laboratory C57BL/6N mice [ 33 , 34 ]. Our results confirm that the chosen ventilator regime avoided undesirable side effects of mechanical ventilation, such as hyperinflation of the lung, hyper- or hypoxia, alkalosis or acidosis, etc.…”

“…In order to explore whether Exo-d-MAPPS treatment managed to improve extracellular acidbase status and gas exchange in CS-exposed mice, blood gas parameters (partial pressure of oxygen in arterial blood (PaO 2 ), partial pressure of carbon dioxide (PaCO 2 ) in arterial blood, oxygen saturation (SaO 2 ), and pH) were analyzed. For this purpose, arterial blood samples were obtained from control and experimental animals and analyzed within a few minutes using a test cartridge blood analysis system (Premier GEM 3500, Instrumentation Laboratory, Bedford, Massachusetts, USA) [29].…”

“…Another technique is oxygen-enhanced MRI. 42,43 Due to its paramagnetic characteristics, oxygen modulates the MRI signal upon inhalation by inducing changes in tissue relaxation. However, the expected changes may be limited in high field strengths.…”
