Assessing the Risk of Birth Defects Associated with Exposure to Fixed-Dose Combined Antituberculous Agents during Pregnancy in Rats

Abstract: Due to the risks of disease progression and transmission to the newborn, treatment of tuberculosis is often pursued during pregnancy and fixed-dose combined antituberculous agents have been found to be beneficial. Unfortunately, there is paucity of data on the safety of the fixed-dose combined antituberculous drugs during pregnancy. This study intends to assess the teratogenic effect of fixed-dose combined antituberculous drugs on the organogenesis stage of fetal development and also investigate the possible r… Show more

“…The first detection of sperm on the plugs marked day D-0 of gestation. Rats have an average gestational period of 21 days ( Awodele et al, 2012 ). Antibiotics suspensions were orally administered to rats on the 13th day of gestation because the first 12 days of rat pregnancy are critical for embryogenesis ( Zalgevičienė, 2010 ; Er, 2013 ).…”

“…In a recent South African study, the use of bedaquiline in pregnancy was linked to an increased incidence of low infant birth weight [105]. Ethionamide has been associated with teratogenicity in some animal models and central nervous system defects in humans [106]. However, there have been case reports where ethionamide use did not lead to similar concerns, and the consensus is that this drug may be used if necessary [107].…”
