Assessment of Bandsharing Values in RAPD-PCR Analysis of Dwarf Cattle of Kerala

Abstract: Random amplified polymorphic DNA (RAPD-PCR) analysis of 56 animals of four different genetic groups of dwarf cattle in Kerala was done as a single step analysis. Bandsharing (BS) values were calculated for animals of each group and between groups as an analytical tool to find out genetic variation among animals. The different factors affecting BS values were estimated using Harvey's Least squares analysis. The effects of genetic group, Guanine-cytosine (GC) content of primer and gel on BS values were found sig… Show more

“…Genetic characterization of native breeds is the first step in prioritization of breeds for conservation. In the recent past, RAPD-PCR approach has been used successfully in numerous studies to reveal informative polymorphism in several species and characterization of genetic variability among breeds of species (Caetano-Anolles et al, 1991;Hunt et al, 1992;Kantanen et al, 1995;Mohd-Azmi et al, 2000;Shilin et al, 2001;Appannavar et al, 2002;Saifi et al, 2004;Suprabha et al, 2005). Technically RAPD assay is simple, rapid, sensitive and low cost method for producing much polymorphic information of the target genome.…”

PCR-based analysis can determine the genetic characteristics of individuals and populations, as highlighted by the substantial GD observed between the two Scapharca populations.…”

“…The Euclidean GDs between two populations of bastard halibut ( Paralichthys olivaceus ) were determined to be 0.018–0.489 from the pooled data for the eight primers used ( Yoon, 2018 ). This technique might help define the diagnostic markers related to the breed, species, genus, and geographic population identification in finfish and crustaceans ( Smith et al, 1997 ; Klinbunga et al, 2000 ; McCormack et al., 2000 ; Yoon & Park, 2002 ; Yoon & Kim, 2004 ; Park et al, 2005 ; Siti Azizah et al, 2005 ), plant ( Bartish et al, 2000 ; Nozaki et al, 2000 ) and cattle ( Suprabha et al, 2005 ).
