Assessment of genetic diversity and phylogenetic relationships of Korean native chicken breeds using microsatellite markers

Abstract: ObjectiveThis study was conducted to investigate the basic information on genetic structure and characteristics of Korean Native chickens (NC) and foreign breeds through the analysis of the pure chicken populations and commercial chicken lines of the Hanhyup Company which are popular in the NC market, using the 20 microsatellite markers.MethodsIn this study, the genetic diversity and phylogenetic relationships of 445 NC from five different breeds (NC, Leghorn [LH], Cornish [CS], Rhode Island Red [RIR], and Han… Show more

“…Several studies, using DNA fingerprinting, polymorphic microsatellite markers, and 600k SNP chip data, found evidence of such genetic introgressions into the KNC since 1960. Based on the previous results, KNC lines are closely related to Cornish, New Hampshire, and RIR breeds and genetically more distant from the white Leghorn breed ( Lee et al., 1996a , b ; Kong et al., 2006 ; Oh et al., 2008 ; Suh et al., 2014 ; Seo et al., 2017 ; Seo et al., 2018 ). “…As the demand for native chicken meat has increased owing to its unique taste and meat quality, the original KNC breeds and commercial breeds have been widely used for diverse crossbreeding since 2000 ( Park et al., 2010 , 2011 ; Kang et al., 2010 ; Lee et al., 2013 , 2014 ). As a result, one of the famous hybrids called “Woorimatdag” produced by the NIAS and “Hanhyup-3-ho” of Hanhyup private company are available on the current market ( Choo and Chung, 2014 ; Seo et al., 2017 ).…”

“…The primers were LEI0258-F: 5′-CAC GCA GCA GAA CTT GGT AAGG-3′ and LEI0258-R: 5′-AGC TGT GCTC AGT CCT CAG TGC -3′ [30,31]. BF2 sequence-based typing (SBT) was performed by sequencing each product obtained from the exon 2 region of the chicken BF2 gene using the forward primer 5′-GCA GAG CTC CAT ACC CTG CGGTA-3′ and the reverse primer 5′-GCC GGT CTG GTT GTA GCG CCG-3′ [32,33]. All primers were synthesized by Bioneer Inc. PCR products were electrophoresed on a 1.5% agarose gel with 0.5 X TBE buffer.…”

“…Microsatellite population studies are more frequently applied on local breeds e.g. in China [7], Oman [8], Korea [9] to clarify origin or position of these breeds relative to other ones. Population study can also be performed within a breed [10] to clarify herd position and highlight those populations which require attention by the management.…”
