Assessment of viability of wharton's jelly mesenchymal stem cells encapsulated in alginate scaffold by WST-8 assay kit

Abstract: Cell encapsulation utilizing biodegradable material has promising outcomes for tissue engineering. From a long time ago, alginate has been generally utilized for drug delivery, cell transplantation and as a scaffold in biomedical applications. The aim of this study was the comparison of cell viability in the presence of two polymerizing ions: Ba2+ and Ca2+ to improvement the quality of alginate scaffold. For this purpose, WJMSCs after three passage were encapsulated in alginate scaffold in the presence of Ba2+… Show more

“…The WST-1 assay is water-soluble, eliminating the need for a separate formazan dissolving step [ 45 , 46 ]. The WST-8 assay was developed from WST-1 and has the advantage that it is less toxic and more sensitive than other types of colorimetric assays [ 46 , 47 ]. The SRB assay is designed to be more sensitive than the MTT assay based on its ability to bind essential amino acid residues to proteins and does not depend on metabolic activities in measuring the viability of living samples [ 48 , 49 ].…”

“…Then, the collected samples are directly placed in culture flasks (explant cultures) or enzymatically digested (collagenase I/II, hyaluronidase, trypsin). In the next step, the processed solution is filtered or centrifuged and transferred into culture dishes and cultured in various media at 37 °C in a 5% CO 2 atmosphere [ 197 , 223 , 224 , 225 ]. Fortunately, the isolation of MSCs from the umbilical cord tissue is much more efficient.…”
