Assignment of the major and minor components of gentamicin for evaluation of batches

Abstract: The antibiotic agent gentamicin sulfate is a mixture of several structurally very similar aminoglycosides. The major components of gentamicin are gentamicin C1, C1a, C2 and C2a. These major components were separated by means of column chromatography. The 1 H and 13 C NMR spectra of the free bases and the corresponding sulfates were assigned using HMQC, HMBC and selective TOCSY experiments. Additionally, the 1 H and 13 C NMR spectra of the sulfates and hydrochlorides, respectively, of the minor components were … Show more

“…GM C2b sulfate (also known as sagamicin and micronomicin) was purchased from Pharm Chemical (Shanghai Lansheng, Shanghai, China). The GM components C1, C1a, and C2/C2a were separated from a commercial sample of GM by column chromatography and analyzed by TLC, CE and 1 H NMR [24]. Paromamine was a gift of the group of Hoogmartens (Leuven, Belgium).…”

“…The spectra of all components of gentamicin were assigned in a previous study [1]. Having this information in hands 1 H NMR spectra can be used to quantify the components in the mixture and quantification results could be confirmed recently by an independent micellar electrokinetic capillary chromatography (MEKC) method [2,3].…”
