Association analysis, genetic diversity and structure analysis of tobacco based on AFLP markers

Abstract: Knowledge in the area of genetic diversity could aid in providing useful information in the selection of material for breeding such as hybridization programs and quantitative trait loci mapping. To this end, 50 Nicotiana tabacum genotypes were genotyped with 21 primer combination of amplified fragment length polymorphism (AFLP). A total of 480 unambiguous DNA fragments and 373 polymorphic bands were produced with an average of 17.76 per primer combination. Also, the results revealed high polymorphic rate varin… Show more

“…During this study, 11 iPBS-retrotransposon markers resulted in an average of 0.33 PIC for 96 Turkish tobacco germplasms. Dadras et al (2014) obtained a mean PIC value of 0.26 using a panel of 50 tobacco genotypes with AFLP markers, which is lower than the value of 0.33 obtained in our study. Lu et al (2013) investigated 330 high-quality DArTseq markers from a panel of 121 tobacco accessions and they resulted in an average PIC value of 0.34, which is close to our resulted PIC value.…”

“…However total soluble sugars per plant was negatively significantly correlated with nicotine percentage as total nicotine alkaloids and chlorides revealing the possibility to improve tobacco quality with low nicotine and chlorides using current population. The observed positive correlation of total soluble sugars with various traits was supported by earlier workers (Maleki et al, 2011;Dadras et al, 2014). This indicates the relative utility of all these traits for selection with respect to sugar content and low nicotine.…”

“…Thus, there is sufficient chance for improvement of these traits and selection based on the phenotypic performance will be reliable and effective from this MAGIC F2 population. Furthermore, moderate to high heritability, GCV and GA% in a mean could be explained by additive gene action and their improvement could be achieved through mass selection in tobacco (Dobhal, 1987;Prasanna and Rao, 1989;Shah et al, 2008;Dadras et al, 2014) in other crops (Ogunbayo et al, 2014;Khatun et al, 2015;Hari et al, 2017).…”

“…Such a demonstration has been represented by some ISSR or SSR association studies on quince (Cydonia oblonga), white savory (Satureja mutica), Coffea canephora and Calophyllum inophyllum, respectively (Ganopoulos et al, 2011;Pawar et al, 2011;Khadivi-Khub et al, 2014;Achar et al, 2015). Based on amplified fragment length polymorphism (AFLP)-PCR, Dadras et al (2014) successfully identified a set of AFLP markers that closely linked to seven growth traits in tobacco. Pereira da Costa et al (2014) reported applying SRAP to identify the DNA loci linked to tomato fruit quality traits.…”
