Several studies have shown a role for the involvement of interleukin (IL)-1 gene cluster polymorphisms in the risk of periodontal diseases. In the present study, we tested polymorphisms, derived from genes of the IL-1 cluster, for association with generalized aggressive periodontitis (GAP) through both allelic association and by constructing a linkage disequilibrium (LD) map of the 2q13-14 disease candidate region. The IL-1RN (VNTR) genotype distribution observed was significantly different in GAP and control subjects (P ¼ 0.019). We also observed some evidence for an association between GAP and the IL-1B þ 3953 polymorphism (P ¼ 0.039). The pattern of association in the region, represented as an LD map, identifies a recombination hot area between the IL-1B þ 3953 and IL-1B À511 polymorphisms. Multilocus modelling of association with disease gives a location for the peak association at the IL-1B þ 3953 marker, although support for the peak is not significant. Haplotype analysis identifies a IL-1B þ 3953 -IL-1B À511 haplotype as having the lowest P-value in the region. Recognition of the presence of a recombination hot area between the IL-1B þ 3953 and IL-1B À511 polymorphisms will have an important bearing on future efforts to develop higher resolution SNP analysis in this region for both this and other diseases for which this cluster is implicated.

ResumoO objetivo deste trabalho foi fazer uma revisão da literatura a respeito do polimorfismo em citoquinas relacionadas ao processo inflamatório periodontal e de como esse fator genético poderia estar ligado ao desencadeamento da doença destrutiva do periodonto. Foram levados em consideração estudos sobre determinadas citoquinas, como: fator de necrose tumoral-alfa (TNFα), interleucina -1-alfa (IL-1α ), IL-1 beta (IL-1β) ou IL-1 receptor antagonista (IL-1RA), IL-2, IL-4 ,IL-6, IL-10, IL-16 e IL-18. A revisão da literatura possibilitou concluirmos que: (1) em algumas citoquinas, como IL-1, IL-2, IL-4, IL-6, IL-10, há variação nos resultados em relação à presença do gene polimórfico e o desenvolvimento da periodontite; (2) alguns genes polimórficos, como os da IL-16 e IL-18, não apresentam relação com a periodontite; (3) existe um enfoque relevante em relação aos grupos étnicos e raciais; (4) o genótipo para IL-1B está associado ao risco para periodontite severa, na população diabética, mas os resultados são contraditórios quando o consumo de cigarros é estudado.Palavras -chave: polimorfismo -citoquinas -doença periodontal; periodontite -fumo;periodontite -diabetes mellitus.

Periodontitis is deined as an inlammatory disease of supporting tissues of teeth characterized by progressive destruction of the periodontal ligament and alveolar bone. Periodontal manifestations of these genetic disorders or syndromes, such as familial and cyclic neutropenias, granulomatous disease, agranulocytosis, Langerhans' cell disease, glycogen storage disease, hypophosphatasia, leucocyte adhesion deiciency, and Papillon-Lefèvre, Chédiak-Higashi, Cohen, Ehlers-Danlos, Marfan, Down, Haim-Munk, and Kindlers syndromes, imitate some types of periodontal diseases. Most of these syndromes have autosomal-recessive characterization and can be seen commonly in consanguineous marriages. Therefore, consanguineous marriages have generally been accepted as having important detrimental efects on ofspring. There is a lot of genetic research about consanguineous marriage and its detrimental efects on ofspring. Although consanguineous marriages are common in the world, the relationship with oral and periodontal diseases has not been thoroughly investigated. We do not have enough of an understanding of the efects of consanguineous marriage on oral and periodontal diseases. In this chapter, previous studies in the literature related to this subject will be investigated and evaluated, and then this research will be related to oral and periodontal diseases. Therefore, this chapter will guide further research. The aim of this chapter is to show the relation between consanguineous marriages and oral-periodontal diseases.
