Milk is one of the basic sources of nutrition. With the increasing incidence of allergies and the spread of knowledge about a healthy lifestyle, consumers are changing their eating habits, which partly leads to the reintroduction of goat's milk into the diet. This study aimed to evaluate and describe the genotypic and allelic structure of Slovak White Shorthaired goats for the CSN1S1 gene. For genotyping, PCR, AS-PCR and PIRA-PCR methods. The He and F IS were used for the description of the diversity of the population. Alleles E and F were the most frequent 0.3710 and 0.2944 respectively followed by B (0.1492) and A (0.1169). The less abundant allele was N with an occurrence of 0.0685. The most abundant genotypes were EF (0.2339), BE (0.1693) AN and EE with an occurrence of 0.1129 both. The White Shorthaired goat breed exhibits high values of heterozygosity (0.7661), polymorphism information content (0.6921), the effective number of alleles (3.7736) and level of possible variability realization (74.1%) for the CSN1S1 gene. The inbreeding level of the population represents a negative value (-0.0355), which indicates enough heterozygotes in the population.

Milk protein polymorphism was analysed to improve the protein content in milk. The present study characterises the CSN1S1 gene and the effect of allelic combinations on milk composition traits in Jamunapari goats. The allelic variants obtained from sodium dodecyl sulfate polyacrylamide gel electrophoresis and polymerase chain reaction–restriction fragment length polymorphism were confirmed by cloning and sequencing. Genetic parameters were obtained from 518 records from 48 sires and 131 dams. The A, B and F alleles were observed in the population and the protein percentage in milk was significantly (P < 0.01) affected by allelic variants. The frequencies of A, B and F alleles were 0.456, 0.503 and 0.041, respectively. The protein content in milk was highest in the goats with AB genotype followed by AA > BB > BF > AF > FF. The goats with AB genotype had a significantly (P < 0.01) higher protein percentage in milk than goats with BF (t = 5.311, df = 113), AF (t = 8.13, df = 123) and FF (t = 9.55, df = 115) genotypes. The direct heritability for protein percentage was 0.441. Parity and season of birth had significant effects (P < 0.05) on the solids‐not‐fat percentage and lactose concentrations. The CSN1S1 AA, AB and BB genotypes should be selected for higher protein content and to improve milk quality and processing traits in Indian goats.

This study was performed to determine polymorphism in the leptin gene (T117C) and investigate the association between genotypes and milk yield in 101 crossbred Anglo-Nubian dairy goats from Awang, Opol, Misamis Oriental and Talay, Dumaguete City, Negros Oriental in the Philippines. Milk yield performance data were standardized to 90 and 140 d milking periods. The goat’s hair follicles were collected to extract genomic DNA. Polymerase chain reaction– restriction fragment length polymorphism (PCR-RFLP) was the method used in genotyping with NmuCI as the restriction enzyme. Statistical analysis was performed following the two-way factorial (2 x 4) in a randomized complete block design (RCBD), where leptin genotypes and parity were used as main factors and farm as the blocking factor. Two genotypes (TT and TC) were obtained. Genotypic frequencies between TT (0.50) and TC (0.50) were evenly distributed, while allele T (0.75) occurred more frequently than allele C (0.25) in the total population studied. Observed heterozygosity is higher than the expected heterozygosity and the genetic variation observed in the overall population was in Hardy-Weinberg equilibrium (p > 0.05). Milk yield traits at 140 d milking period in crossbred Anglo-Nubian dairy goats were significantly influenced by polymorphisms in leptin (T117C). Goats having TT genotype had superior milk yield performance (140dADMY: 0.90L ± 0.058L, 140dTMP: 124.65L ± 8.51L) than those with TC genotype (140dADMY: 0.78L ± 0.051L, 140dTMP: 107.90L ± 7.21L). Further studies should be conducted in other goat breeds to validate associations of leptin polymorphism with milk production and in other economically important traits.
