Association of the HNK-1 epitope with 5′-nucleotidase from Torpedo marmorata (electric ray) electric organ

Abstract: 5'-Nucleotidase isolated from the electric organ of the electric ray (Torpedo marmorata) has a molecular mass of 62 kDa and, on two-dimensional electrophoresis, separates into up to 13 isoforms within a pI range of 5.9-6.7. The N-terminal sequence data show a 71% identity over 17 amino acids with that previously published for the rat liver enzyme. All forms of 5'-nucleotidase are recognized by the HNK-1 monoclonal antibody. HNK-1 immunoreactivity is found at the surface of the Schwann-cell processes covering t… Show more

“…in [201,246]). In addition, in Torpedo electric organ [246] and in developing kitten visual cortex [247], the HNK-1 epitope, a sulfated, glucuronic acid-containing carbohydrate epitope, that has been found in a variety of cell adhesion molecules, has been identified. The glycosylation pattern presumably varies not only between species but also between tissues.…”

“…The isolation of 5'-nucleotidase from bovine adrenal medulla, rat brain and porcine brain capillaries (Weber et al, 1987) was carried out essentially by the same method, except that the detergent sulfobetain-14 (1 YO), instead of Triton X-300 ( 5 % ) and deoxycholate (2'/0), was used to solubilize the membranes. N-terminal amino acid residues from the Torpedo enzyme were determined by direct protein sequencing using automated Edman degradation (Vogel et al, 1991). Polyclonal antibodies were obtained by immunization of rabbits with the purified protein.…”

“…The function of ecto-5'-nucleotidase has generally been allocated to the dephosphorylation of extracellular mononucleotides. Interestingly, 5'-nucleotidase isolated from the electric ray electric organ contains the HNK-1 epitope (Vogel et al, 1991), which has been identified as glucuronyl3-sulfate. This epitope is associated with a number of surface-located membrane proteins which are involved in cell adhesion or cell recognition.…”

“…15 CD57 1 T cells fail to proliferate after antigen-specific stimulation in vitro and are highly vulnerable to activation-induced apoptosis. 16,17 Furthermore, it was demonstrated that replicative senescence of T cells is better defined by the expression of CD57 than by a lack of CD28 expression, as determined by measurement of the T-cell receptor excision circle content and of the proliferative ability of T cells. 17 Notably, while CD57 1 T cells have been shown to be associated with various inflammatory diseases, [18][19][20][21][22] the pathogenic roles of CD57 1 T cells have yet to be elucidated in coronary artery disease.…”
