Association Studies of Variants in the Genes Involved in Pancreatic β-Cell Function in Type 2 Diabetes in Japanese Subjects

Abstract: Because impaired insulin secretion is characteristic of type 2 diabetes in Asians, including Japanese, the genes involved in pancreatic ␤-cell function are candidate susceptibility genes for type 2 diabetes. We examined the association of variants in genes encoding several transcription factors (TCF1, TCF2, HNF4A, ISL1, IPF1, NEUROG3, PAX6, NKX2-2, NKX6 -1, and NEUROD1) and genes encoding the ATP-sensitive K ؉ channel subunits Kir6.2 (KCNJ11) and SUR1 (ABCC8) with type 2 diabetes in a Japanese cohort of 2,834 … Show more

“…1 Several studies in various ethnic populations have shown association of polymorphisms in genes involved in b-cell development and function with T2D and associated phenotypes. [2][3][4][5][6][7] Importantly, genome-wide association studies have also deciphered associations with loci predominantly involved in b-cell function. 8 Indians have the highest incidence and prevalence of T2D worldwide.…”

“…1-4 However, a large-scale association study of these genes has not been carried out in the Chinese Han population; and there was much inconsistency in results in East Asian studies before. [5][6][7][8][9] In this study, 1912 unrelated type 2 diabetes patients and 2041 normal controls (Supplementary methods) were recruited from Shanghai, China, by measuring their clinical characteristics (Supplementary Table 1). Both the SNPs (single nucleotide polymorphisms) mentioned above were genotyped by TaqMan SNP Genotyping Assays on an ABI7900 system (Applied Biosystems, Foster City, CA, USA).…”
