Associations of PCR markers with freezing tolerance and photosynthetic acclimation to cold in winter barley

Abstract: Associations of PCR markers with freezing tolerance and acclimation of photosynthetic apparatus to cold were tested on 28 winter barley cultivars and advanced breeding forms to select alleles for practical application in marker assisted selection (MAS). We found significant associations between freezing tolerance evaluated with field-laboratory method (FLM) and markers located on 5H chromosome in region of gene Fr-H2 (bin9-10: Xbmag812, Xmwg2230) and region of gene Fr-H1 (bin11: Xmwg514, HvBM5, Xmwg644). “…The CG marker can thus be used not only for selection of GH types in winter x spring crosses, but also for fast routine selection of frost tolerant genotypes (Akar et al 2009). Similar indications came from the work of Rapacz et al (2010) that found variation in the promoter region of Vrn-H1 (HvBM5) directly connected with freezing tolerance of plants partially de-acclimated in the field.…”

“…PCRs were as described previously (Rapacz et al. ). Digestion of selected products with restriction enzymes was carried out in 10 or 15 μl volumes containing 2× concentrated buffer recommended for the respective restriction enzymes (Fermentas, Vilnius, Lithuania) and 10 or 5 μl of PCR product, respectively.…”

“…A less simple genetic model was obtained from controlled environment experiments, where nine freezing tolerance QTLs, distributed across four of the seven barley chromosomes, were identified (Tuberosa et al 1997). More recently, the loci Fr-H1 and Fr-H2, both involved in the determination of freezing tolerance, were mapped to the long arm of barley chromosome 5H (Skinner et al 2006;Francia et al 2007), and DNA-based genetic markers for these loci have been developed since then (Akar et al 2009;Rapacz et al 2010).…”
