Atorvastatin Inhibits Inflammatory Response, Attenuates Lipid Deposition, and Improves the Stability of Vulnerable Atherosclerotic Plaques by Modulating Autophagy

Abstract: Atherosclerosis is a chronic disease comprising intima malfunction and arterial inflammation. Recent studies have demonstrated that autophagy could inhibit inflammatory response in atherosclerosis and exert subsequent atheroprotective effects. Our previous study also demonstrated the role of autophagy in the inhibition of inflammation by atorvastatin in vitro. Therefore, in the present study, we aimed to determine whether atorvastatin could upregulate autophagy to inhibit inflammatory cytokines secretion, lipi… Show more

“…2A). Among the autophagy-associated proteins, the conversion of LC3-I to LC3-II has been reported to be widely used as a marker of autophagic cell death (19). The present study did not indicate any significant changes in the ratio of LC3-II/LC3-I between control and ICA-treated groups (Fig.…”

“…Previous studies have demonstrated that basic autophagy deficiency by specific Atg5 knockout could accelerate atherosclerotic . Moreover, our previous studies have suggested that up‐regulated autophagy level by atorvastatin could inhibit inflammatory cytokines secretion and lipid accumulation and even suppress atherosclerosis progression both in vitro and in vivo . However, the beneficial effects of autophagy on vulnerable plaques and the potential mechanism are still little known.…”

“…In addition, the NF-κB inhibitor decreased the expression of inflammatory mediators in inflammatory cells. It was suggested that the activation of the NF-κB pathway was involved in the regulation of the NLRP3 inflammasome [108,109]. erefore, atorvastatin plays an anti-inflammatory role by inhibiting the PMA-induced THP-1 monocyte via the TLR4/MyD88/NF-KB pathway.…”
