ATP and luciferase assays to determine the rate of drug action in in vitro cultures of Plasmodium falciparum

Abstract: BackgroundKnowledge of the rate of action of compounds against cultured malaria parasites is required to determine the optimal time-points for drug mode of action studies, as well as to predict likely in vivo parasite clearance rates in order to select optimal hit compounds for further development. In this study, changes in parasite ATP levels and transgenic luciferase reporter activity were explored as means to detect drug-induced stress in cultured parasites.MethodsIn vitro cultures of Plasmodium falciparum … Show more

“…The IC 50 concentrations of six out of seven drugs tested (chloroquine, artemisinin, dihydroartemisinin, atovaquone, lumefantrine and quinine) were not significantly different from WT (Fig. S3, Tables S2 and S3) and were comparable to previously published data (Duraisingh et al ., , de Monbrison et al ., , Quashie et al ., , Bacon et al ., , Walsh et al ., , Hayat et al ., , Khan et al ., , Chaorattanakawee et al ., , Hasenkamp et al ., ). Only for mefloquine we detected a small but significant decrease in sensitivity for the three mutants; however, all IC 50 values are well above previously reported values.…”

“…Atipik morfolojiye sahip türlerin görülmesinin sebepleri arasında olası genotipik değişik-likler, yetersiz/uygunsuz tedavi, hastadan örneğin tedavi sırasında alınmış olması gibi faktörler yer almaktadır 13 . Atipik morfoloji nedeniyle Plasmodium tür ayrımının yapılamadığı durumlarda antijen varlığını gösteren hızlı testler ve/veya moleküler yön-temlere başvurulmalıdır.…”
