Attachment of Rod-Like (BAR) Proteins and Membrane Shape

Abstract: Previous studies have shown that cellular function depends on rod-like membrane proteins, among them Bin/Amphiphysin/Rvs (BAR) proteins may curve the membrane leading to physiologically important membrane invaginations and membrane protrusions. The membrane shaping induced by BAR proteins has a major role in various biological processes such as cell motility and cell growth. Different models of binding of BAR domains to the lipid bilayer are described. The binding includes hydrophobic insertion loops and elect… Show more

“…20 Third, changes in local membrane curvature followed by wrapping of NPs and their agglomerates may lead to lipid depletion from the bilayer, and thus altered membrane integrity and/ or formation of pores or holes. 16,22,[53][54][55][56][61][62][63][64] In the case of GUVs, we propose that the most likely mechanism causing GUVs to burst is adhesion of CAadsorbed CoFe 2 O 4 NPs and smaller NP agglomerates onto membranes, resulting in alteration of the global membrane curvature and, finally, bursting. The POPC lipids used in our study have an externally situated lipid polar head (the trimethylammonium group) that is positively charged and faces the aqueous glucose solution.…”

“…Recently, the anisotropic nature of curvature has received increasing attention theoretically. The classical Canham-Helfrich curvature free energy [32,33] has been extended to anisotropic curvatures [34][35][36]. To simplify the interactions, the protein and membrane underneath it have been often modeled together as an undeformable object with a fixed curved shape such as a point-like object with an anisotropic curvature [37,38] and a bent elliptical surface [39].…”

“…However, another explanation consistent with the above findings is that the majority of nanoparticles are created after blood sampling, and accordingly the results reflect the properties and composition of blood cells and plasma. For example, membrane and plasma constituents may affect membrane curvature, [47][48][49] which is the basic mechanism underlying the budding and vesiculation of membranes. Controlled manipulation of the size of the cell fragments (nanoparticles) could be advantageous, as avoiding production of fragments of the size of immune complexes (mean diameter approximately 150 nm) may avoid problems with artefacts when measuring the nanoparticle concentration by flow cytometry.…”
