Plant resistance characteristics are closely related to changes in the activities of self-defense enzymes after infection. Despite significant differences in the resistance of different lily (Lilium sp.) cultivars to leaf blight (Botrytis elliptica), few studies of their resistance physiology exist. This study explored changes in the resistance-related enzyme activity of several lily cultivars after leaf blight inoculation. Seven oriental lily cultivars (Lilium hybrids) with obvious differences in resistance were used as experimental materials. After inoculation with B. elliptica, the activities of four defense enzymes, superoxide dismutase (SOD), catalase (CAT), phenylalanine ammonia-lyase (PAL), and peroxidase (POD), were determined according to the light absorption values at different wavelengths after their reactions. The results showed that the activities of SOD and CAT differed between the highly resistant and highly susceptible hybrids. Before inoculation, SOD activity was relatively low in all cultivars. However, after inoculation, the SOD activity increased sharply in the resistant cultivars. In the moderately resistant cultivars, the SOD activity did not change drastically. In the susceptible cultivars, the SOD activity initially showed slight increases or decreases and then increased. CAT activity showed reactions similar to those of SOD. Some changes in PAL and POD activity occurred after inoculation, but no significant correlations were present between these trends and resistance characteristics. In addition, no significant changes in enzyme activities were found in the control plants of the seven cultivars during the testing period. Overall, the resistance of Lilium oriental hybrids to B. elliptica is related to SOD and CAT activity but does not show much of a relationship with PAL and POD activity. Studying the physiological metabolic pathways of SOD and CAT appears to be an important direction in research to elucidate resistance to B. elliptica in Lilium oriental hybrids.

Marker-assisted selection (MAS) is implemented to improve Camellia oleifera yield and fruit attributes for meeting China’s increased demand for new varieties development. We conducted three-generational (G0, G1, and G2) hybridization (diallel mating) and selection experiment and used Sequence-Related Amplified Polymorphism (SRAP). SRAP markers to investigate their utility in a MAS framework. The utilized SRAP markers were instrumental in hybrid authenticity and the identification of matroclinal or patroclinal inheritance presence, thus guiding mating pair selection and direction (their role as male or females). Across the studied 3 generations, estimates of genetic diversity parameters showed steady increase with percentage increase of ((G0 to G1 and G1 to G2) 9.25 and 9.05: observed number of alleles; 3.12 and 7.80: means effective number of alleles; 12.35 and 22.34: Nei‘s gene diversity; and 14.21 and 21.77: Shannon‘s index), indicating lack of diversity reduction associated with selection. Estimates of genetic distance and their correlation with heterosis were useful in guiding selection of mating pairs for achieving the desired yield and fruit attributes (fruit diameter, height, weight, and index, peel thickness, number of seeds per fruit, seed weight per fruit, and seed rate). Most yield and fruit attributes exhibited high broad-sense heritability with increasing trend over generation intervals, indicating the increased potential of hybrid breeding for this species.

Fire Blight Susceptibility in Lilium spp. Correlates to Sensitivity to Botrytis elliptica Secreted Cell Death Inducing Compounds

Fire blight represents a widespread disease in Lilium spp. and is caused by the necrotrophic Ascomycete Botrytis elliptica. There are >100 Lilium species that fall into distinct phylogenetic groups and these have been used to generate the contemporary commercial genotypes. It is known among lily breeders and growers that different groups of lilies differ in susceptibility to fire blight, but the genetic basis and mechanisms of susceptibility to fire blight are unresolved. The aim of this study was to quantify differences in fire blight susceptibility between plant genotypes and differences in virulence between fungal isolates. To this end we inoculated, in four biological replicates over 2 years, a set of 12 B. elliptica isolates on a panel of 18 lily genotypes representing seven Lilium hybrid groups. A wide spectrum of variation in symptom severity was observed in different isolate-genotype combinations. There was a good correlation between the lesion diameters on leaves and flowers of the Lilium genotypes, although the flowers generally showed faster expanding lesions. It was earlier postulated that B. elliptica pathogenicity on lily is conferred by secreted proteins that induce programmed cell death in lily cells. We selected two aggressive isolates and one mild isolate and collected culture filtrate (CF) samples to compare the cell death inducing activity of their secreted compounds in lily. After leaf infiltration of the CFs, variation was observed in cell death responses between the diverse lilies. The severity of cell death responses upon infiltration of the fungal CF observed among the diverse Lilium hybrid groups correlated well to their fire blight susceptibility. These results support the hypothesis that susceptibility to fire blight in lily is mediated by their sensitivity to B. elliptica effector proteins in a quantitative manner. Cell death-inducing proteins may provide an attractive tool to predict fire blight susceptibility in lily breeding programs.
