Autophagic activation in vitrified–warmed mouse oocytes

Abstract: Vitrification involves the use of cryoprotectants (CPAs) and liquid nitrogen (LN 2 ), which may cause osmotic damage and cryoinjury to oocytes. Autophagy is widely recognized as a survival or response mechanism elicited by various environmental and cellular stressors. However, the induction of autophagy in vitrified-warmed oocytes has not been examined. In this work, we investigated whether the vitrification-warming process induces autophagy in mouse oocytes. Metaphase II (MII) oocytes that were vitrified and … Show more

“…Beclin1 is a mammalian homolog of yeast autophagy-related gene 6 ( Atg6 ) and is essential for autophagy. Induction or phosphorylation of Beclin1 is required to initiate autophagosome formation through the activation of class III phosphoinositide 3-kinase (PI3K) 24 . In GFP dsRNA-injected control oocyte, Beclin1 proteins were observed as small spots and were distributed evenly in the ooplasm ( Fig.…”

“…Consistent with this, autophagy activation has been reported in vitrified-warmed oocytes. 31 Also, rapamycin treatment has been shown to induce autophagy, improving in vitro maturation of porcine oocytes. 32 In addition to cytokinetic failure, we found a few misaligned chromosomes in BECN1 knockdown oocytes.…”

“…Oocytes that survived the thawing process (∼98%) were subjected to lipid extraction. In our routine laboratory procedures, vitrified-warmed mouse oocytes showed average of 62% fertilization rate and ∼50% developmental rate to the blastocyst stage [36].…”
