Autophagy inhibition impairs the epithelial-mesenchymal transition and enhances cisplatin sensitivity in nasopharyngeal carcinoma

Abstract: Drug resistance restricts the efficacy of cisplatin in the treatment of nasopharyngeal carcinoma (NPC). Increasing evidence indicates that autophagy and the epithelial-mesenchymal transition (EMT) participate in cancer progression and drug sensitivity. The aim of the present study was to investigate the function of autophagy and EMT in cisplatin treatment, and to reveal the underlying impact of autophagy on the EMT process in NPC. Transmission electron microscopy assays and western blot analyses confirmed that… Show more

“…Manipulation of autophagy has also been demonstrated to prevent an EMT-like phenotype and associated metastasis/invasion in a number of cancer cell lines and tissues, including breast, colorectal, pancreatic and ovarian cancers [42][43][44][45][46][47][48] . In hepatocellular carcinoma, autophagy inhibition was not shown to induce EMT and had no effect on migration or invasion [43] .…”

“…Anti-cancer drugs have increasingly been utilised in combination with autophagy inhibitors. When cisplatin was used in combination with autophagy inhibition, this increased cytotoxicity in cells [48,104] Similarly, the effects of 5-FU are augmented in colon cancer when treated with autophagy inhibitor hydroxychloroquine [105] .…”

“…Therefore, it is important to find novel treatment strategies for HNC. Uncontrolled invasion and metastasis contribute to this poor prognosis, and recent study results have suggested that epithelial-mesenchymal transition (EMT) serves an essential role in cancer cell metastasis, invasion, radiotherapy resistance, drug resistance, immune evasion and the cancer stem-cell phenotype (4,5). Previous studies have demonstrated that a number of critical biomarkers, including E-cadherin, N-cadherin and vimentin, are involved in EMT (5,6).…”

“…To further to examine the contributions of autophagy induced by SMI-4a treatment, the autophagy inhibitor CQ was used to pharmacologically inhibit SMI-4a-induced autophagy. CQ functions to block lysosomal acidification and autophagosome degradation [23]. Our findings indicate that inhibition of autophagy with CQ could remarkably potentiate SMI-4a-induced antitumour activity in vitro and in vivo, indicating the cytoprotective role of autophagy in SMI-4a therapy for melanoma.…”
