Autophagy-Mediated Inflammatory Cytokine Secretion in Sporadic ALS Patient iPSC-Derived Astrocytes

Abstract: Background. Astrocytes can be involved in motor neuron toxicity in amyotrophic lateral sclerosis (ALS) induced by noncell autonomous effects, and inflammatory cytokines may play the main role in mediating this process. However, the etiology of aberrant cytokine secretion is unclear. The present study assessed possible involvement of the mTOR-autophagy pathway in aberrant cytokine secretion by ALS patient iPSC-derived astrocytes. Method and Results. PBMCs from sporadic ALS patients and control subjects were rep… Show more

“…Therefore, we suggest that SIRT3 is involved in regulating Xyl‐B‐activated autophagy. Autophagy was found to be associated with inflammatory responses, especially the release of IL‐6 and TNF‐α inflammatory mediators 39,40 . Our data suggest that Xyl‐B has regulatory effects on inflammatory factors both in vivo and in vitro, while pretreatment with the autophagy inhibitor 3‐MA reduced the regulatory effects of Xyl‐B on intracellular inflammatory factors.…”

“…We further explored the effect of OGD/R on autophagy-related proteins. Beclin1 is an important autophagy regulatory gene that recruits autophagy key protein to autophagic vesicle precursor, forming a complex with Vps34 and Vps15; all of these provide an indispensable condition for the formation of autophagic vesicle . LC3 includes LC3I and LC3II, and LC3II is a marker molecule of autophagic vesicles and serve as a read-out of an autophagic vesicles number .…”

“…The derived cells were fixed with 4% paraformaldehyde (Sigma, St. Louis, MO, USA) in DPBS (Invitrogen, Carlsbad, CA, USA) for immunofluorescence staining with the markers nestin and SOX2 [SRY (sex-determining region Y)-box 2] to identify the NSCs. Subsequently, NSCs were cultured in NSC amplification medium [ 30 ]. Passage 3 were used in our experiments.…”
