Autoradiographic determination of brain pH following middle cerebral artery occlusion in the rat.

Abstract: SUMMARY A method of quantitative autoradiography using M C-labelled 5,5-dimethyl-2,4-oxazolidinedione I4 C-DMO to evaluate the local changes in brain pH after ischemia is described. In normal control raU the calculated tissue pH values in gray matter were slightly lower than those in white matter, and there was no significant difference in the calculated pH among the various structures in cortical and subcortical gray matter. Four hours after a left middle cerebral artery (MCA) occlusion, marked reductions in … Show more

“…The LCpH values reported here in the control rats are similar to those published previously by Kobatake et al (1984) in the same animal model and assumed by Mabe et aI. (1983) to prevail in the intracellular space following short -duration isch emia.…”

“…In thiamine deficiency, focal cerebral acidosis, which is most likely due to lactate accumulation, can result in a pH of <6.5 in various cerebral struc tures (Hakim, 1984) despite normal or even ele vated LCBF (Hakim, 1986). To address this point more specifically, LCpH and LCBF are now being determined at earlier intervals following occlusion.…”

“…2 implies prior knowledge of pHe and Ceo Kobatake et al (1984) assumed that pHe = pH p and that C e = C p o Though this assumption may be valid for tumor tissue if the blood-brain barrier (BBB) is deficient, it is prob ably not valid for normal gray and white matter. If…”

“…The DMO technique has been used in normal rats and after occlusion of the middle cerebral artery (Kobatake et al, 1984;Hakim, 1986) but not, to our knowledge, under the circumstances of the present study. Since the methods appear to be valid after middle cerebral artery occlusion, an insult more likely than the conditions of the present study to have an adverse effect on the assumptions that un derlie the DMO technique, we believe that this is a warranted extension of the method.…”
