Avena phytochrome a overexpressed in transgenic tobacco seedlings differentially affects red/far-red reversible and very-low-fluence responses (cotyledon unfolding) during de-etiolation

Abstract: Abstract. Etiolated seedlings of tobacco (Nicotiana tabacum L.) were exposed to single light pulses predicted to establish different proportions of phytochrome in its far-red absorbing form (Pfr/P). The angle between the cotyledons was compared in wild-type and transgenic seedling overexpressing Arena phytochrome A over the range of both very low-fluence responses (VLFR) and low-fluence responses (LFR). The unfolding of the cotyledons increased linearly for 24 h after the light pulse. At this time the Pfr/P-re… Show more

“…6). In dicotyledonous plants, a similar effect of phyA overexpression has been interpreted as the result of an exacerbated HIR in light-grown seedlings, impairing the response to changes in R/FR caused by supplementary FR (McCormac et al 1991;Casal and Sfinchez 1994). Third, rice transgenics were able to respond to canopy shade light.…”

“…Third, rice transgenics were able to respond to canopy shade light. Light-grown tobacco plants overexpressing phyA respond to natural shade light and other experimental reductions in R/FR ratio unless caused by supplementary FR (Casal and Sfinchez 1994).…”

“…Work by Nagy et al (1993) and Casal et al (1994) can be interpreted to indicate that phyA is the phytochrome that entrains the circadian oscillator. A very-low-fluence response can phase shift the circadian cycling of Lhcb mRNA in T .…”

“…Thus, under Re or Rp, the mode of action of phyA can be defined as 'multiple VLFR'. There are other systems, such as cotyledon unfolding in etiolated tobacco (Casal, Sanchez & Vierstra 1994), mesocotyl growth in rice (Clough et al 1995) and seed germination in Arabidopsis (Botto et al 1996), where phyA (overexpressed or endogenous) selectively induces VLFR. Therefore, phyA operates via two different action modes: HIR (under FRc) and VLFR.…”
