Avian Influenza A Virus Pandemic Preparedness and Vaccine Development

Abstract: Influenza A viruses can infect a wide range of hosts, creating opportunities for zoonotic transmission, i.e., transmission from animals to humans, and placing the human population at constant risk of potential pandemics. In the last hundred years, four influenza A virus pandemics have had a devastating effect, especially the 1918 influenza pandemic that took the lives of at least 40 million people. There is a constant risk that currently circulating avian influenza A viruses (e.g., H5N1, H7N9) will cause a new… Show more

“…The highly pathogenic avian influenza (HPAI) viruses of the A/goose/Guangdong/1/1996 (GsGd) lineage have become a major global health concern soon after its emergence in 1996 in China (Xu, Subbarao, & N. J., & Guo, Y., ). Since then, the GsGD lineage of HPAI H5N1 virus has continued to spread globally killing millions of poultry with occasional spill overs to wild birds and mammals, including humans (de Vries, Herfst, & Richard, ). As of 1 November 2018, HPAI H5N1 virus has been responsible for 860 confirmed human infections from 16 countries claiming 454 lives (WHO, ).…”

“…Highly pathogenic avian influenza (HPAI) H7N9 virus remains global concerns. Current strategies for developing and manufacturing these vaccines are time consuming and somewhat limited in the ability to generate vaccines quickly [15]. Furthermore, conventional influenza vaccines utilizing the HA and NA of influenza viruses have safety and production issues [16].…”

“…Sera were isolated from blood samples collected at day 13 and 28 after the initial immunization, and stored 18 at -20 ℃ until use. Avidin-biotin system (ABS)-enzyme-linked immunosorbent assay (ELISA) was 19 conducted to measure HA-specific IgG titers, as described previously [11]. Briefly, 96-well microplates 20 (Costar, Corning, NY, USA) were coated with 2 ÎŒg / mL of recombinant HA protein, and incubated 21 overnight at 4 ℃.…”
