Avocado Juice Prevents the Formation of Trypsin Amyloid-Like Fibrils in Aqueous Ethanol

Abstract: In this work fruit and vegetable juices were analyzed for their ability to prevent the aggregation of trypsin using turbidity measurement. Fruit and vegetable juices are capable of inhibiting the aggregation of PMS-trypsin in aqueous ethanol. Among the juices examined, avocado was found to be the most effective. Choline bitartrate was investigated for its ability to inhibit the fibrillation of PMS-trypsin. We have found that avocado juice and choline bitartrate have an inhibitory effect on the formation of try… Show more

“…Ethanol (EtOH) is the most commonly used alcohol in beverages available in the market with the highest percentage varying from 3–50 % (v/v) [13] . Aggregation of Trypsin is triggered in the presence of 60 % EtOH as reported by Kotormán and co‐workers [14–17] . Different mutations and inactivation of genes due to autodigestion of pancreatic Trypsin, triggered by alcohol consumption, serve as the active precursor of pancreatitis and pancreatic cancer [18] .…”

“…[13] Aggregation of Trypsin is triggered in the presence of 60 % EtOH as reported by Kotormán and co-workers. [14][15][16][17] Different mutations and inactivation of genes due to autodigestion of pancreatic Trypsin, triggered by alcohol consumption, serve as the active precursor of pancreatitis and pancreatic cancer. [18] Pglycoprotein expressed by more than 95 % of pancreatic cancer cells excludes the chemotherapeutics from entering, thereby making the treatment of pancreatic cancer more difficult.…”

“…The presence of a mild solvent causes an increase in the β-sheet conformation in proteins (Furkan et al 2016;Simon et al 2012), thus the organic solvents can be used to prepare amyloid fibrils (Nematilay et al 2014;Kotormán et al 2015). Amyloid growth detection is generally performed by measuring the turbidity of the solution (Zhao et al 2016;Kasi and Kotormán 2019b).…”

“…Date palm fruits ( Phoenix dactylifera ) have been reported in studies for their potential biological capacity in nephroprotective, hepatoprotective, and anticancer activity [ 35 , 36 , 37 ]. Recently, the inhibitory effect of avocado juice against trypsin aggregation has been reported: a formation process associated with several neurological diseases [ 38 ]. Similarly, other reports identified the beneficial role of berries (blackberry and blueberry) with regards to the obstruction of the central nervous system and cognitive deficit [ 39 , 40 ].…”
