Avoiding Drug-Drug Interactions

Abstract: Background: Drugs may be prescribed in combinations causing drug-drug interactions (DDI) and adverse drug reactions (ADR), resulting in hospital care. Methods: To provide prescribers of drug therapy with a better knowledge of individuals’ current drug therapy, governments have started to collect prescribing data. Results: The data on individuals’ dispensed drugs is available for prescribers, pharmacists, and the registered individuals in Sweden and Denmark. The information has not yet come to an extensive use … Show more

“…Nevertheless, other studies are necessary to analyze methotrexate concentration at a given time point but also the area under the curve of methotrexate with and without PPI, in order to not simply categorize outcomes as delayed elimination or not delayed. To avoid such potential drug-drug interactions, reliable information about drugs needs to be available in databases accessible to all health professionals [28]. Prescribers and pharmacists have to know the individual’s current drug therapy, especially if PPI or methotrexate is taken by the patient.…”

“…It should be mentioned that irrational combinations do not necessarily involve drug-drug interactions; therefore this paper will touch upon potential interactions. According to B.Astrand, 17% to 23% of drug combinations are associated with such a risk, and only from 6% to 8% of them actually result in an interaction (Astrand 2009). However, a number of factors, such as a narrow therapeutic index of many psychoactive drugs (Spina et al 2016, Hiemke et al 2018, predominantly hepatic metabolism involving the cytochrome P450 system (Guo et al 2012, de Leon andSpina 2018), and antipsychotic polytherapy (use of several antipsychotic drugs) (Tranulis et al 2008, Correll et al 2009, Misawa et al 2011, can increase the occurrence of drugdrug interactions in the treatment of psychiatric inpatients.…”

“…The extensive distribution and diverse physiologic functions of Pgp leads us to consider that this protein may have an important role in the process of absorption, distribution and elimination of several drugs. Because the participation of Pgp and estrogenic hormones in drug-drug interactions has not been widely studied, further investigations need to be done to prevent the decrease in therapeutic effect or adverse reactions [28] .…”
