B‐1 cells produce insulin and abrogate experimental streptozotocin‐induced diabetes

Abstract: The participation of B-1 cells in a murine model of spontaneous diabetes has been recently reported. Here, we describe the role of B-1 cells in streptozotocin (STZ) induced diabetes in mice. We demonstrated that XID (B-1 cell-deficient) mice are more susceptible to STZ treatment than WT mice, as evidenced by their higher blood glucose level in response to STZ. Unexpectedly, the XID mice that were i.p. transferred with purified B-1 cells, either before or after the STZ treatment, did not develop diabetes. These… Show more

“…Phenotypical cell analysis included the quantification of macrophages, B and T lymphocytes. Pre-B-1 CDP is a stage of differentiation of B-1 in B-1 CDP, which is described in infection and also diabetes mice model [ 20 , 21 ]. There was a significant decrease of peritoneal B-1, B-2 and CD4 + T lymphocytes of DM and DM-Infected mice than non-diabetic ( Infected and Uninfected ) mice ( Fig 3 ), showing that DM condition decreased the immune response.…”

“…Type 1 DM was induced by intra-peritoneal administration of STZ at 50 mg/kg/day, during 5 consecutive days [ 20 , 21 ]. Ten days after the end of the treatment, 10 μl of blood was collected from the tail of each DM animal.…”

“…Non-adherent cells were discarded and RPMI-1640 supplemented with 10% FCS (R10) was added to the adherent fraction. To obtain B-1 CDP, APerC from BALB/c mice were cultured and the enriched B-1 cells in the floating medium were collected from the third day [57]. Cultures of 0.5 × 10 6 cells/well were re-suspended in R10 and re-cultured under the same conditions as described above.…”

“…XID mice were adoptively transferred with B-2 cells from the spleen of BALB/c mice [46]. After tissue dissociation, lysis of red blood cells and centrifugation, a pellet was incubated with anti-CD16/32 for blocking Fc receptors, for 15 minutes at 4°C.…”
