Contribution of CD4+ and CD8+ T lymphocyte subsets to the cytokine secretion patterns induced in mice during sensitization to contact and respiratory chemical allergens

Abstract: SUMMARYChemical allergens of different types, those that cause in humans allergic contact dermatitis or occupational asthma, induce in mice divergent immune responses characteristic, respectively, of T-helper 1 (Th1)-and Th2-type cell activation. Such responses are associated with the development of different cytokine secretion patterns by draining lymph node cells (LNC), such that contact allergens stimulate vigorous interferon-g (IFN-g) production, but little secretion of the Th2 cytokines interleukin-4 and … Show more

“…There was no evidence for induction of Tc2 cells. However, IFN-g was produced in formaldehyde-sensitized mice by CD4 π Th1 cells and in DNCB-sensitized mice by both CD4 π Th1 cells and CD8 π Tc1 cells (40). These data illustrate that in response to some skin sensitizing chemicals, both CD4 π and CD8 π cell responses are induced, but that this is not necessarily a universal rule.…”

“…The converse type 2 cytokine secretion profile was provoked by the chemical respiratory allergen trimellitic anhydride (TMA) (11,12). These polarized cytokine phenotypes take time to mature, with LNC isolated 3 days after initiation of exposure expressing both type 1 and type 2 cytokines following treatment with either chemical contact or respiratory allergens (13)(14)(15). These data suggest that the molecular signature of chemical allergens may be being misread by the immune system as pathogens, resulting in Th1-or Th2-type polarization.…”

“…However, until now, there is no validated method to assess the potentials of chemicals to induce respiratory sensitization. Several groups of researchers have attempted to create a model to test chemicals' potential to induce respiratory sensitization [7][8][9][10][11]. In these attempts, the researchers use different animal models, including, rats, mice, and guinea pigs and assess lung ventilatory as well as immunological determinants that could predict respiratory sensitization potentials.…”
