B1b Lymphocytes Confer T Cell-Independent Long-Lasting Immunity

Abstract: Many microbial pathogens employ antigenic variation as a strategy to evade the immune system, posing a challenge in vaccine development. To understand the requirements for immunity against such pathogens, we studied Borrelia hermsii, a relapsing fever bacterium. We found that mice deficient in T, follicular B, marginal zone B, or B1a lymphocytes resolved B. hersmii bacteremia and became resistant to reinfection. The resolution of bacteremia coincided with an expansion and persistence of B1b lymphocytes, and pu… Show more

“…B. hermsii-specific IgM induced by the bacteremia is capable of clearing the vmp-expressing spirochete population from the host (4). B1b cells are one source of B. hermsii-specific IgM that can control low dose bacteremia associated with attenuated (high passage) B. hermsii variants, as has been shown using xid mice, which are deficient in the B1b cell subset (6). However, Ab produced by these cells is less effective in clearing virulent (low passage) B. hermsii (5).…”

“…These findings underscore the importance of pathogen-specific IgM in the clearance of Borrelia spirochetes and suggest that B cell subsets other than follicular B cells contribute to protective immunity against infection with these organisms. In this regard, the B1b cell subset has been shown to be a source of protective Ab against B. hermsii, but only after priming by infection (6). Naive B1b cells, in contrast, produce Ab that can clear attenuated (high passage) B. hermsii, but are unable to clear the first episode of spirochetemia in primary infection of mice with virulent (low passage) B. hermsii (6).…”

“…[5][6][7]. However, the precise contribution of T-cell-dependent and TI pathways to IgA production is not known. An important contribution of B-1 cells to TI responses has been suggested [6,8,9]. Peritoneal cavity (PEC) B-1 cells have also been claimed to contribute significantly to IgA-producing plasma cell (PC) Eur.…”

“…Nevertheless, there is increasing evidence of a broader role for MZ B cells in both T-independent (TI) and T-dependent (TD) immune responses (3,4). It has recently been documented that B-1b cells are mostly responsible for the adaptive immune response to TI Ag and exert a memory function (5,6). Both B-1a and MZ B cells express polyreactive specificities with low affinities to a broad range of Ags and are thus responsible for the production of natural Abs, thereby contributing to innate immunity (1,6).…”
