Aim: To determine the prevalence of Listeria monocytogenes and associated risk factors among beef operations (cow‐calf and feedlot) in central and southern California. Methods and Results: A repeated cross‐sectional study where faecal and environmental samples were collected from 50 operations three times a year at different seasons was carried out. Samples were tested for presence of L. monocytogenes using a combination of enrichment and polymerase chain reaction tests. Data on putative risk factors were also collected. Listeria monocytogenes was detected in faecal samples from cows, calves and other animals on calf‐cow operations at proportions of 3·1%, 3·75% and 2·5%, respectively. The organism was detected in 5·3% of cut‐grass, 5·3% of soil, 14·3% of irrigation ditches, 3·1% of the ponds and 6·5% of water troughs samples. Listeria monocytogenes was less common in faecal (0·3%) and soil (0·75%) samples collected from feedlots. Conclusions: Listeria monocytogenes was present at a higher proportion among cow‐calf operations than feedlots. There was no significant seasonal variation in the occurrence of this pathogen within the two types of operations. Significance and Impact of the Study: If risk mitigation strategies were implemented to reduce the public health risk these should focus in cow‐calf operations.

Short-wave W-energy (100 p,W/cmZ) decreased the number of List&u monocytogenes on Tryptose Agar (TA) ca. seven orders of magnitude in 4 min. Age of culture (48 vs 24 hr) did not alter the sensitivity of Listeria to this W treatment. Increasing the intensity of irradiation (550 vs 100 pW/cm2) increased the rate at which L. monocytogenes was inactivated. Dry rather than moist cells of L. monocytogenes were most resistant to irradiation. In general, inactivation of Lisferiu with short-wave W-energy yielded sigmoidal survivor curves. The population of Listeria on TA plates was not affected with long-wave Wirradiation.

Listeria monocytogenes 4b surface protein extract was used in an immunoblot assay to analyze the antibody profile in sera from 40 healthy urban workers (U group), 40 healthy slaughterhouse workers (W group) and four healthy carriers with positive L. monocytogenes 4b feces culture (positive controls). In addition, pooled rabbit sera, before and after immunization with L. monocytogenes 4b whole-cell suspension, were analyzed against L. monocytogenes 4b surface protein extract in order to determine the specific L. monocytogenes 4b antibody pattern. The degree of similarity (S) between such a pattern and each of those obtained with serum samples from the three subject groups was assessed. For U and W group sera, mean S values were 24.3 ± 13.5 and 32.8 ± 14.3, respectively. An S value greater than 65, corresponding to mean S U value ± 3 standard deviation, was considered as an indicator of a healthy carrier. Thus, the estimated healthy carrier percentages found in U and W groups were 2.5 and 5%, respectively. The proposed immunoblot assay may prove a useful tool for epidemiological surveys to determine whether a healthy person is a L. monocytogenes 4b carrier.
