Baculovirus Display of Varicella–Zoster Virus Glycoprotein E Induces Robust Humoral and Cellular Immune Responses in Mice

Abstract: Varicella–zoster virus (VZV) is the causative agent of varicella and herpes zoster (HZ) and can pose a significant challenge to human health globally. The initial VZV infection—more common in children—causes a self-limiting chicken pox. However, in later life, the latent VZV can become reactivated in these patients, causing HZ and postherpetic neuralgia (PHN), a serious and painful complication. VZV glycoprotein E (gE) has been developed into a licensed subunit vaccine against HZ (Shingrix). However, its effic… Show more

“…Immunization with rBV displaying the rhoptry protein ROP4 protected mice from a lethal challenge with T. gondii, by inducing mucosal, cellular and humoral immunity 23 . The ability of BV-based vaccines to induce strong immune responses and protection by displaying heterologous antigens was also demonstrated for Plasmodium (PfCSP) 35 , and several viruses, including a recent work with SARS-CoV2 22,[36][37][38] . All these studies, however, were carried out displaying the heterologous antigens at BV surface.…”

“…Immunization with recombinant baculovirus displaying the rhoptry protein ROP4 protects mice from lethal challenge with T. gondii by inducing mucosal, cellular, and humoral immunity (Yoon et al, 2021). The ability of BV-based vaccines to induce strong immune responses and protection by displaying heterologous antigens has also been demonstrated for Plasmodium (Yoshida et al, 2018) and several viruses, including a recent work on SARS-CoV2 (Abe et al, 2003;Premanand et al, 2018;Wei et al, 2021;Xue et al, 2022). However, all of these studies have been carried out displaying heterologous antigens on the BV surface.…”

“…Fusion of proteins to full GP64 results in chimeric proteins that possess all the signals required for transport to the membrane, maturation, and incorporation into the budded virus. Using this strategy, it has been possible to expose large proteins such as GP120 of HIV [ 44 ] and ROP4 of Toxoplasma gondii [ 45 ].…”
