Baicalin-loaded PEGylated lipid nanoparticles: characterization, pharmacokinetics, and protective effects on acute myocardial ischemia in rats

Abstract: The results of the present study imply that PEG-NLC could be the biocompatible carriers for heart-targeted drug delivery to improve myocardial ischemia.

“…The underlying mechanisms of ischemia-reperfusion injury include free radical damage, calcium overload, leukocyte activation, and microvascular damage [810]. Myocardial ischemia-reperfusion injury will produce a large number of free radicals, leading oxidation, crosslinking, denaturation, and degradation of DNA, RNA, protein, and polysaccharide molecules and eventually causing the myocardial cell apoptosis and death [11]. Therefore, in the process of myocardial ischemia reperfusion, increased production of free radicals should be halted.…”

“…This indicated that the drug was rapidly eliminated after i.v. injection in piglets and was much slower than the rate reported in mice (Jin et al, ) but faster than for rats (Li et al, ; Xing et al, ; Yang et al, ; Zhang et al, ) and rabbits (Wei et al, ). The V d was 1.63 ± 0.23 L/kg indicating that baicalin was widely distributed into body tissues.…”

“…12,13,15 Recent advances in nanoscale drug carriers for MI therapy include liposomes (containing PEGylated liposomes), core-shell hybrid liposomal vesicles, lipid nanoparticles, etc. [16][17][18][19] In this study, we designed Sch B loaded PEG-modified solid lipid nanoparticles (SLNs) for MI treatment.…”

“…42 This confirms that the PEGylation of nanoparticles could improve the plasma half-life of the carriers and achieve sustained-release of the loaded drugs. 16 PEG was reported to have the ability to improve the surface hydrophilicity of lipid particles and prevent the absorption of lipoproteins and opsonins effectively. 22 Therefore, the PEG ligands present over the carriers could avoid the recognition of the reticuloendothelial system and prolong the circulation time, thus bringing about a persistent therapeutic effect.…”
