Transportation is of high risk and cannot be avoided during the productive practice of Lates calcarifer fry. To achieve an optimal transporting protocol, the changes of physico‐chemical indexes and immune genes expression of fry after transportation were evaluated. The fish were simulated transported at the density of 7 kg/m3 for different time (T1 = 4 h, T2 = 8 h, T3 = 12 h, T4 = 16 h, T5 = 20 h, T6 = 24 h) and at three densities (D1 = 14 kg/m3, D2 = 21 kg/m3, D3 = 28 kg/m3) for 24 h. With the intensive stress, a decrease in pH and an increase in NH4+‐N were observed. At transporting density experiment, the cortisol of every group except T3 had a significant difference with the control. ALT, AST and ACP activity were all close to a rise first and then decline trend. IL‐8, TNF and IFN‐γ1 were activated to promote inflammation at different degrees of transportation pressure, while the tendency of L10 and TGF‐β1 was similar. CRP also had no significant increase under different transporting durations stress. HSP70 undertook main repair work as expression level of it increased gradually. Complement did not reach its release standard at duration experiment expect for T3 group of C3. However, density experiment had no obviously positive effects on physico‐chemical indexes and the relative expression of immune genes. The results indicated that when the transporting time exceeded 8 h at the density of 7 kg/m3, significant immune response started happening to fish. The transporting density stress has stimulated the fries but the immune response was still under the inhibited state owing to the ammonia toxicity reduction by much lower pH.

Black body syndrome causes mass mortality in barramundi (Lates calcarifer) in Asia mariculture. This study aimed to evaluate the efficacy of bivalent vaccine derived from local isolate for black body syndrome (BBS) prevention on the Barramundi. The bacteria used in the bivalent vaccine were Pseudomonas stutzeri and Vibrio harveyi in a ratio of 50:50. Barramundi, 5‒6 cm in length, was intraperitoneally injected with bivalent vaccine and phosphate buffer saline (PBS). After a 21-day vaccination, fish was injected with pathogenic bacteria P. stutzeri, V. harveyi, the combination of P. stutzeri and V. harveyi with a dose of bacteria 107 CFU/fish. We observed relative percent survival (RPS), mortality, blood profile, antibody level, lysozyme activity, and histopathology of vaccinated fish. The result showed that vaccinated fish had higher antibody levels and lysozyme activity than control treatment (P>0.05). Vaccinated fish had RPS of 80.00%, 64.29%, 57.69%, after challenged test with P. stutzeri, V. harveyi, and combination of P. stutzeri and V. harveyi, respectively. Hemoglobin and hematocrit were not significantly different (P>0.05). However, the erythrocytes, leucocytes, and phagocytic activity were higher compared to there were higher erythrocytes, leucocytes, and phagocytic activity compared to control (P<0.05). As well as antibody level and lysozyme activity of vaccinated fish higher than control (P<0.05). In conclusion, the bivalent vaccine of P. stutzeri and V. harveyi could protect barramundi seed from BBS infection. Tujuan dari penelitian ini adalah untuk menguji efikasi vaksin bivalen untuk penanggulangan black body syndrome pada benih ikan kakap putih. Bakteri yang digunakan untuk membuat vaksin bivalen yaitu Pseudomonas stutzeri dan Vibrio harveyi dengan rasio 50:50. Ikan kakap putih berukuran 5‒6 cm diinjeksi vaksin bivalen dan phosphat buffer saline secara intraperitoneal. Setelah 21 hari pemeliharaan, benih diuji tantang dengan bakteri patogen P. stutzeri, V. harveyi, campuran P. stutzeri dan V. harveyi dengan kepadatan bakteri sebesar 107 CFU/ikan. Parameter yang diamati meliputi relative percent survival (RPS), gambaran darah, titer antibodi, aktifitas lisosim, dan gambaran histopatologi benih kakap putih. Hasil penelitian menunjukkan bahwa ikan yang divaksin memiliki nilai titer antibodi dan aktifitas lisosim lebih tinggi dibandingkan kontrol (P<0.05). Ikan yang divaksin memilki RPS 80.00%, 64.29%, 57.69%, setelah diuji tantang dengan P. stutzeri, V. harveyi, campuran P. stutzeri dan V. harveyi. Nilai hemoglobin dan hematokrit tidak berbeda nyata (P>0.05). Titer antibodi dan aktifitas lisosim ikan yang divaksin lebih tinggi dari kontrol (P<0.05). Seabass, Lates calcarifer (Bloch 1790) is an euryhaline and a popular fish in south and southeast Asian countries due to its higher growth rate, giant size, excellent taste and higher market demand. It is locally known as Bhetki or Koral and available around the year in the estuarine and coastal regions of Bangladesh. In many countries (e.g. Australia, India, Malaysia, Philippines, Thailand etc.), the species is artificially propagated and commercially cultured. The breeding technology has not yet been developed in Bangladesh. Now a days Bangladesh is progressing day by day in freshwater aquaculture but it is far behind in case of coastal and mariculture. To get the mass seed production of fry/fingerlings of seabass, we need to develop the artificial breeding technology of the said species. There are only a few studies published on biology, aquaculture, brood development and artificial breeding activities of seabass in Bangladesh where investigators used live feed (Tilapia) and small indigenous fish species (SIFS) on brood development in captivity but none could adapt to formulated feed on seabass aquaculture. Some researchers made attempt to develop seabass broodstock for artificial breeding purpose, but none can success in captive condition in Bangladesh. Only one breeding trial conducted by stripping method using wild brood, but the development of the fertilized eggs ceased attaining up to the neurola stage, Res. Agric., Livest.
