Behavioral effects of estrogen receptor gene disruption in male mice

Abstract: Gonadal steroid hormones regulate sexually dimorphic development of brain functions and behaviors. Their nuclear receptors offer the opportunity to relate molecular events in neurons to simple instinctive mammalian behaviors. We have determined the role of estrogen receptor (ER) activation by endogenous estrogen in the development of male-typical behaviors by the use of transgenic estrogenreceptor-deficient (ERKO) mice. Surprisingly, in spite of the fact that they are infertile, ERKO mice showed normal motivat… Show more

“…Dose-dependent and centrally mediated weight reduction follows oestradiol intake, 15 while its impact on testosterone is more complex, mediating through negative feedback and low LH levels. 4,15 While an ejaculatory dysfunction was precipitated in mice by oestrogen alpha-receptor inactivity, 16 high oestradiol intake for 12 weeks compromised arousal, desire (mounting) as well as sexual performance (intromission) in one of our treatment groups Table 1 Serum levels (ng/ml) of total E 2 and T in the different experimental groups. E 2 -T ratio gives the hormonal imbalance arising from oral treatment with E 2…”

“…The masculinization and defeminization of male sexual behavior in rodents are traditionally believed to rely on the conversion of androgens to estrogens and the subsequent activation of ERs during the perinatal period, a hypothesis most recently supported by data from mice deficient in either aromatase or ERs (Ogawa et al, 2000;Matsumoto et al, 2003;Ogawa et al, 1997). However, newborn male rats administered an aromatase inhibitor retain masculine sexual behaviors (Dominguez-Salazar et al, 2002), while genetically produced AR knockout mice (ARKO) show less masculine sexual behavior (Sato et al, 2004), suggesting that ARs are also involved in male sexual behavior.…”

“…Accordingly, copulation and other sexually motivated behaviors are severely impaired in ERαKO males (Eddy et al, 1996;McDevitt et al, 2007;Ogawa et al, 1997;Wersinger and Rissman, 2000;Wersinger et al, 1997). We have utilized the ERα −/AA mouse model to investigate the relative contributions of classical and non-classical ERα signaling in male sexual behavior.…”
