Benzimidazo[2,1-b]quinazolin-12-ones. New class of potent immunosuppressive compounds

Abstract: Immunosuppressive Compoundswas added, and the flask was tightly closed and left at room temp.At the end of the required reaction time the solvent was removed under reduced pressure, and the product was purified as described in Table I.Method C.-In a flask as above and using a condenser, d-pilocarpine (0.02 mole) was dissolved in Me»CO (10 ml) with constant stirring while heating to about 50°. A freshly prepd soln of 0.025 mole of the halo organic reagent in 10 ml of Me2CO was then slowly added and the mixt was… Show more

“…Like other benzimidazole derivatives [5,13,15,17,23], ZIMET 3164 showed a dosedependent suppression of humoral antibody formation in the test systems employed. Both primary and secondary immune response were affected and, in contrast to cyclophosphamide [4], the immunological memory, too.…”

“…Benzimidazoquinazoline dertivatives were first described for their antitumoral activities (Lunn et al, 1971;El-Sherbeny, 2000; Via et al, 2000) including In the model described here, pentoxifylline reached only a 77% inhibition of PMA/LPS-mediated TNF-α production when used at 2 mM final concentration, as compared with >90% inhibition of the 6-Phenyl-benzimidazo [1,2-c]quinazoline derivative (G1) tested at 100 μM. Pentoxifylline is a well-known vasoactive drug with proven clinical efficacy in various circulatory disorders, and is also a commonly used drug during sepsis (Staubach et al, 1998) and to improve the outcome from sepsis in premature infants (Kirsch and Giroir, 2000).…”

“…Previous reports described benzothiazoloand benzimidazoquinazoline derivatives as cytotoxic compounds with potential antitumoral activity (Lunn et al, 1971;ElSherbeny, 2000;Via et al, 2000), and particularly, there is one report that describes the antitumoral activity of benzimidazo [1,2-c]quinazolines (Brana et al, 1994).…”
