Benzimidazole-2-Phenyl-Carboxamides as Dual-Target Inhibitors of BVDV Entry and Replication

Abstract: Bovine viral diarrhea virus (BVDV), also known as Pestivirus A, causes severe infection mostly in cattle, but also in pigs, sheep and goats, causing huge economical losses on agricultural farms every year. The infections are actually controlled by isolation of persistently infected animals and vaccination, but no antivirals are currently available to control the spread of BVDV on farms. BVDV binds the host cell using envelope protein E2, which has only recently been targeted in the research of a potent and eff… Show more

“…It may be that the PI animals produced continuously expel viruses to the external environment, leading to the expansion of infection. Some literature points out that there is no antiviral drug to prevent the spread of BVDV in the farm at present, and the spread of the virus can only be prevented by isolating PI animals or vaccinating ( 177 ). Therefore, BVDV detection should be done well in the breeding season to reduce the production of PI animals and control the spread of BVDV from the source.…”

“…However, the antiviral activities of these old drugs are not very satisfactory. Benzimidazole derivatives display a variety of biological activities in many diseases through various mechanisms, such as antiviral [ 28 , 29 ], antimicrobial [ 30 , 31 ], antiproliferative [ 32 , 33 ], and anticancer activities [ 34 , 35 ]. Benzimidazole derivatives are special lead compounds for antiviral drugs, attracting interest from many chemists [ 18 , 36 , 37 , 38 , 39 ].…”

“…Albeit vaccines are effective against BVDV, and cover BVDV infections, additional therapy is required to eradicate BVDV infections [ 7 ]. With this aim, a promising option is the use of non-nucleoside inhibitors (NNIs), that specifically target the active form of BVDV RdRp, which is differentiated by the positioning of linker and hood ( Figure 1 A,B) [ 4 , 8 ]. Following this route, some selective anti-BVDV inhibitors have been discovered [ 2 , 5 , 8 , 9 , 10 , 11 , 12 , 13 , 14 , 15 , 16 ].…”

However, the molecular recognition of BVDV RdRp with allosteric NNIs has not been extensively studied, as no co-crystallized NNI-RdRp has been published to our knowledge.…”
