Berberine Induces Cell Apoptosis through Cytochrome C/Apoptotic Protease-Activating Factor 1/Caspase-3 and Apoptosis Inducing Factor Pathway in Mouse Insulinoma Cells

Abstract: High concentration (5 and 10 μmol/L) of berberine could induce the apoptosis of MIN6 cells through cytochrome C/Apaf-1/caspase-3 and apoptosis inducing factor (AIF) pathway.

“…(shown in sup.1). Our group has already conducted research on the functions and working mechanisms of the components, such as berberine, curcumin, ginsenoside and Corni Fructus [2629]. In addition, the results from other researchers have also demonstrated anti-diabetic effect of some active ingredients of JTXK granule [30].…”

“…The activated downstream caspases lead to cell apoptosis by cleaving specific substrates [ 23 ]. Caspase 3, a member of the caspase family which has been studied relatively thoroughly, is considered a key protease in apoptosis [ 24 ]. After being activated, it triggers a downstream cascade reaction to induce apoptosis.…”

“…Cyt-c and deoxyadenosine triphosphate (dATP) interact to form the apoptosome. Activation of caspase-3 via its cleavage further inhibits the expression of poly (adenosine diphosphate-ribose) polymerase, thus triggering the apoptotic cascade ( 42 , 43 ). Furthermore, CypD, a cyclosporin A-binding protein located in the mitochondrial matrix, is a component and a key regulator of the mPTP to maintain mitochondrial integrity.…”
