.beta. Subunit of rat liver mitochondrial ATP synthase: cDNA cloning, amino acid sequence, expression in Escherichia coli and structural relationship to adenylate kinase

Abstract: The amino acid sequence of all but a few N-terminal residues of the beta subunit of rat liver ATP synthase has been determined from cDNA clones. Rat liver F1-beta is shown to contain 17 amino acid differences from that reported for F1-beta of bovine heart, 2 differences of which involve differences in charge. This may account in part for the observation that bovine heart F1 binds nucleotides with much greater affinity than the rat liver enzyme. Rat liver F1-beta also contains homologous regions with another nu… Show more

“…Adenine nucleotide translocase isoform 1 (ANT 1 ) mRNA levels were detected using a 1.4-kb ANT 1 cDNA fragment cloned from a human skeletal muscle library (ATCC, Rockville, Maryland) (23,35). ␤F 1 -ATPase mRNA levels were detected using a 1.8-kb ␤F 1 -ATPase cDNA fragment cloned from a human HeLa cell line library (ATCC) (4,37). An inducible heat shock protein (HSP70-1) mRNA level was detected by a 1.7-kb cDNA fragment cloned from human hippocampus (ATCC) (17,21).…”

“…2B) as follows. Most mass peaks corresponded closely (<0.1% mass difference) to the masses of tryptic peptides (17 limit peptides, 4 partials) predicted by direct translation of the Fl-13-subunit cDNA sequence [20,21]. The remaining 10 mass peaks differed from predicted Fl-13-subunit tryptic peptides by 16 Da or 32 Da, suggesting the possibility of methionine oxidation [22].…”

“…Together, these data characterized the Fl-~-subunit as a (macroscopically) low affinity and moderate capacity calcium-binding protein. [20,21,23]) is represented by the identity ratio, with an ambiguous call (in parentheses; the expected residue is given) excluded. Initial sequencing yields were 5-12 pmol.…”
