Betasatellite-encoded βC1 protein regulates helper virus accumulation by interfering with the ATP hydrolysis activity of geminivirus-encoded replication initiator protein

Abstract: Geminivirus–betasatellite disease complexes are an epidemic threat to the majority of economically important crops across the world. Plant virus satellites including betasatellites are maintained by their associated helper virus. Geminivirus–betasatellites influence viral pathogenesis by substantially increasing or decreasing their helper virus accumulation. In the present study, we attempted to understand the mechanistic details of the geminivirus–betasatellite interaction. Here, we used tomato leaf curl Guja… Show more

“…A pGBKT7-P53 and pGADT7-TAg plasmid combination was used as positive control, while a pGBKT7 and pGADT7 vector combination was used as the negative control. The interaction was tested by growing the yeast co-transformants on synthetic screening media lacking Leu, Trp, and His and containing 1 mM 3-amino-1,2,4-triazole and X-α-Gal at 30°C for 4 days ( Gnanasekaran et al., 2019 ; Gnanasekaran et al., 2023 ).…”

“…However, the CLCuB βC1 gene , in combination with bipartite tomato leaf curl New Delhi virus (ToLCNDV), replaces the movement function of viral DNA B by transporting the viral DNA A from the nucleus (the site of virus replication) to the plasmodesmata to facilitate intra- and intercellular movement of the viral DNA A [ 39 ]. In another betasatellite (tomato leaf curl Patna betasatellite) and non-cognate monopartite begomovirus (tomato leaf curl Gujarat virus) interaction, the βC1 protein influences viral pathogenesis by substantially reducing the helper virus accumulation by interfering with the ATP hydrolysis activity of the helper virus Rep protein, as the C-terminal region of βC1 protein interacts with the C-terminus of the Rep (RepC) protein coded by the virus [ 40 ]. Variations in begomovirus–betasatellite interactions have also been shown for cassava-infecting begomoviruses [ 41 ].…”
