Bienzymatic Biosensor for Malic Acid Based on Malate Dehydrogenase and Transaminase Immobilized onto a Glassy Carbon Powder/Carbon Nanotubes/Nad+ Composite Electrode

Abstract: A biosensor for malic acid detection in fruit juices, based on malate dehydrogenase (MDH) glutamate oxaloacetate transaminase (GOT) in a powder vitreous carbon, carbon nanotubes and mineral oil, composite matrix has been developed. By employing amperometry at a constant potential of 0.4 V (vs. Ag/AgCl), an enzymatic reaction promoted by these species in a support solution Mc Ilvaine buffer (phosphate/citrate pH 10.0) with glutamic acid was generated, causing the oxidation of electro active species, that was st… Show more

“…MD simulation is a potent method to study the structure, dynamics, and energetics of the biological systems in the diverse experimental (pressure, temperature, and pH) conditions (Johnson et al, 2009;B. Li et al, 2018;Moradi et al, 2020;Schröder, 2017). Precisely designed MD simulations can frequently intercept the wrong interpretations of the obtained experimental results.…”

“…Biosensors are defined as the analytical tools that have an extensive application in the diverse scientific fields, such as healthcare, clinical diagnostics, pharmaceuticals, food quality control, and environmental monitoring (Bahadır & Sezgintürk, 2015; Cao et al, 2014; Kaçar et al, 2017; Mundaca‐Uribe et al, 2017). A biosensor comprises three basic components, including the bioreceptor, transducer, and signal processing segments.…”

“…MDH enzymes with a preference for NAD + are classified as members of Enzyme Commission (EC) class 1.1.1.37, whereas the plant and algal enzymes with a preference for NADP + are members of EC 1.1.1.82. It is the NAD +preferring enzymes of EC 1.1.1.37 that have been used exclusively for malate biosensors (Giménez-Gómez et al, 2017, Blaedel and Engstrom, 1980, Mizutani et al, 1991, Yoshioka et al, 1992, Silber et al, 1994, Cho et al, 1995, Prodromidis et al, 1996, Gajovic et al, 1997, Katrlı ḱ et al, 1999, Miertuš et al, 1998, Maines et al, 2000, Manzoli et al, 2004, Mazzei et al, 2007, Arvinte et al, 2008, Arvinte et al, 2009, Gamella et al, 2010, Monošík et al, 2012a, Vargas et al, 2016, Mundaca-Uribe et al, 2017, Röhlen et al, 2017.
