Binding Mode and Structure–Activity Relationships of ITE as an Aryl Hydrocarbon Receptor (AhR) Agonist

Abstract: Discovered as a modulator of the toxic response to environmental pollutants, aryl hydrocarbon receptor (AhR) has recently gained attention for its involvement in various physiological and pathological pathways. AhR is a ligand-dependent transcription factor activated by a large array of chemical compounds, which include metabolites of l-tryptophan (l-Trp) catabolism as endogenous ligands of the receptor. Among these, 2-(1'H-indole-3'-carbonyl)thiazole-4-carboxylic acid methyl ester (ITE) has attracted interest… Show more

“…This point is particularly important given the growing interest in AhR research on identifying novel classes of potent, non-toxic AhR ligands for use in various therapeutic settings (Stockinger 2009). Moreover, these observations raise concerns about potential off target immune toxicities associated with the use of ITE based immune therapies (Abron et al 2018;Dolciami et al 2018;Hao and Whitelaw 2013;Nugent et al 2013;Quintana et al 2010b).…”

“…Mouse embryonic fibroblasts (MEFs) were obtained according to the guidelines of the University of Perugia Ethical Committee and the European Communities Council Directive 2010/63/EU. MEFs and Ahr −/− MEFs were prepared as described (14). Briefly, pregnant mice were sacrificed at 13.5 days post-coitum (dpc) by cervical dislocation.…”

“…XRs are also extremely involved in regulating general physiology since they can also transcribe for genes involved in immune regulation, cell metabolism, energy homeostasis (Figure 1). Families of XRs may bind a very large family of unrelated ligands by direct or indirect binding (27,28). XRs may include the pregnane X receptor (PXR), the AhR, the constitutive androstane receptor (CAR), and the peroxisome proliferator-activated receptors (PPARs) (Figure 1).…”
