Binding of bromocresol green and bromocresol purple to albumin in hemodialysis patients

Abstract: In renal insufficiency, the BCP/BCG ratio of serum albumin is affected by multiple factors: next to carbamylation, uremic toxins (total PCS) and α1-acid glycoprotein also play a role.

“…Serum albumin levels were measured conferring the bromocresol green dye-binding method on a Roche Modular DP analyser (Roche Diagnostics) (Delanghe et al 2018). Cholesterol levels were measured with the CHOD-PAP method (Barzehkar et al 2016).…”

“…Confounders comprise carbamylation, 2 uremic toxins (e.g., para-cresyl sulfate, 3 and a 2 -macroglobulin concentration. 4 The altered plasma protein spectrum in nephrotic syndrome (higher a 2 -macroglobulin-albumin ratio) highlights the analytical failure of the dye binding assays. The acute phase protein a 1 -acid glycoprotein 3 affects albumin test results when using bromocresol-based dyes.…”

“…3 In these patients, the performance of BCP methods seems sufficient and would therefore be "fit for intended use." Admittedly, the binding of BCP to albumin is influenced by protein carbamylation 2 and uremic toxins, 4 which has been shown in dialysis patients. The most important lesson is that clinicians should be aware of the pitfalls of chromogenic assays and an adequate alternative measurement method should be used when necessary.…”

“…The effective use of 3,3 -dimethyl-5,5 -dibromophenolsulfonephthalein (BP) in technologies is expanding . It includes sensitive gas sensors [27,28], electrochemical DNA biosensors to the antitumor drug [39], highly efficient extraction of heavy metals from industrial effluents by adsorption on a modified zeolite surface [40], lysine decarboxylase analysis [41], the adsorption of hydrophobic molecules on the membrane of erythroleukemia cells [42], and evaluation of the properties of serum albumin [38,[43][44][45][46][47][48] and extracellular lactase [49]. It is noteworthy that many applications are based on the formation of dissimilar associates between the anionic form of sulfonephthalein and the analyte.…”
