Bio-protective effects of homologous disaccharides on biological macromolecules

Abstract: In this contribution the effects of the homologous disaccharides trehalose and sucrose on both water and hydrated lysozyme dynamics are considered by determining the mean square displacement (MSD) from elastic incoherent neutron scattering (EINS) experiments. The self-distribution function (SDF) procedure is applied to the data collected, by use of IN13 and IN10 spectrometers (Institute Laue Langevin, France), on trehalose and sucrose aqueous mixtures (at a concentration corresponding to 19 water molecules per… Show more

“…It is also of interest to examine the normalization procedures most frequently applied both on the measured elastic scattered intensity, and on the extracted MSD [31,42]. It is well known that the measured scattering law, which is a function of Q, represents the number of elastically diffused neutrons within a given solid angle.…”

“…To be effective in preservation, the medium should effectively reduce the local, fast motions which are probably precursors for protein denaturation or other deactivation processes [46]. In this respect, the reduction of water dynamics, as observed in binary systems [47], could be the mechanism for the invoked suppression of short scale motions.…”

“…Neutron scattering can be used to probe most of these observables. Elastic and quasielastic neutron scattering can access the pico-to nanosecond dynamics (Benedetto and Kearley 2016;Magazù et al 2008Magazù et al , 2009Magazù et al , 2010Magazù et al , 2012Bee 1988;Volino 1978). Neutron spin-echo can be used to investigate slower relaxation processes (Mezei 1972) and to probe structural fluctuations (Nagao 2009;Woodka et al 2012).…”
