Bioactivity and mechanisms of flavonoids in decreasing insulin resistance

Abstract: Flavonoids are ubiquitous compounds in nature and are found in many Chinese herbal medicines. Due to their biological activity, flavonoids show potential for decreasing insulin resistance (IR), thereby delaying the progression of diabetes and accompanying metabolic syndromes. This review focuses on the mechanisms of flavonoids decreasing IR: (1) the interaction between flavonoids and target proteins of the insulin signalling pathway; (2) bioactivities of flavonoids, such as anti-inflammatory, lipid-lowering an… Show more

“…[20] Flavonoids, being the most abundantly occurring and extensively studied among the polyphenols, demonstrate considerable potential in treating NAFLD due to their inherent safety, widespread availability, and diverse biological activities. [21][22][23] The flavonoids identified in this study, namely, quercetin, kaempferol, and naringenin, offer promising advantages in NAFLD treatment through various mechanisms, which is consistent with findings from other studies. [24] Many in vivo and in vitro experimental evidence have identified various molecular mechanisms of quercetin in preventing the occurrence and progress of NAFLD, including antioxidant stress, anti-inflammatory, improving fatty acid metabolism, and regulating gut microbiota and bile acid metabolism.…”

“…Therefore, the consumption of flavonoids as nutraceuticals requires more clinical trials to demonstrate their clinical value in metabolic syndrome treatments. While flavonoid compounds have demonstrated beneficial impacts on insulin resistance, plasma glucose reduction, the enhancement of the lipids profile, and cardiovascular protection in animal models and in vitro experiments, further clinical trials and in vivo experiments are required to confirm their efficacy [55].…”

“…It is an illness with a complicated etiology that affects several body organs. Flavonoids' wide range of pharmacological effects have made them a popular research subject for natural hypoglycemic medications [152]. Fisetin was discovered to be able to suppress the expression of the genes phosphoenolpyruvate carboxykinase (PEPCK) and glucose-6-phosphatase (G-6-Pase), which are the key enzymes of hepatic gluconeogenesis, in order to maintain glucose homeostasis and improve insulin resistance.…”
