Bioavailability Improvement Strategies for Icariin and Its Derivates: A Review

Abstract: In recent years, there has been considerable interest in icariin (ICA) and its derivates, icariside II (ICS) and icaritin (ICT), due to their wide range of potential applications in preventing cancer, cardiovascular disease, osteoporosis, delaying the effects of Alzheimer’s disease, treating erectile dysfunction, etc. However, their poor water solubility and membrane permeability, resulting in low bioavailability, dampens their potential beneficial effects. In this regard, several strategies have been develope… Show more

“…Small amounts (0.25%) of water-soluble polymers and organic salts such as sodium salicylate, hydroxypropyl methylcellulose, sodium carboxymethyl cellulose, polyvinylpyrrolidone, polyethylene glycol 4000, sodium dodecyl sulfate, hexadimethrine bromide, tris(hydroxymethyl)aminomethane, sodium acetate or benzalkonium chloride were added to the complexation reaction. After work up and HPLC analysis, it was found that the complexation with addition of sodium dodecyl sulfate could increase icariin’s water solubility to 13.09 mg/mL, which is the best result obtained so far and is 654 times higher than that of the icariin ( Szabó et al, 2022 ). The inclusion rate and yield of icariin inclusion compound were obtained as 1.8% and 78.12% respectively, and the product was used for inclusion complex confirmation and in vitro and in vivo pharmacokinetic studies.…”

“…Notably, the impact of polyphenols in preventing drug resistance appears encouraging in oncology therapy [ 14 ]. Icariin is one of the main active ingredients of the Chinese botanical drug Epimedium, a well-researched anti-cancer agent [ 76 ]. Icariin induces apoptosis in TMX-resistant breast cancer cells and inhibits autophagy; thus, it is an ideal sensitizing agent.…”

“…Icariin (ICA), an active component extracted from Epimedium species, has broad biological application prospects [ 2 ]. Numerous studies have shown that ICA and its metabolites can improve neuronal survival and function by reducing the production of extracellular amyloid beta peptide (Aβ) [ 3 ] and intracellular neurofibrillary tangles (NFTs) and inhibiting phosphodiesterase-5 activity [ 4 ].…”
