Biocatalytic Synthesis of Quercetin 3‐O‐Glucoside‐7‐O‐Rhamnoside by Metabolic Engineering of Escherichia coli

Abstract: You are what you eat: Quercetin 3‐O‐glucoside‐7‐O‐rhamnoside is a rare natural product with potential biological importance. We describe a strategy for the regioselective synthesis of this bisglycoside by feeding quercetin 3‐O‐glucoside, a naturally abundant precursor, to metabolically engineered Escherichia coli.

“…To establish a relationship between the effects of TMX on root-nodule numbers at the first trifoliate stage and root isoflavones, levels of isoflavones in root tissue were quantified according to Roepke and Bozzo (2013) with slight modifications. Briefly, roots of soybean seedlings at the first trifoliate stage were washed under tap water and gently blotted.…”

“…These flavonol-O-diglycosides contain a combination of rhamnose, glucose, and arabinose at the 3 and 7 hydroxyl groups of flavonols such as kaempferol and quercetin (YonekuraSakakibara et al 2008). E. coli was also used to synthesize these flavonol diglycosides (Kim et al 2013a;Roepke and Bozzo 2013). Quercetin-3-O-glucoside-7-O-rhamnoside was synthesized by two approaches.…”
