Biochemical Investigations of Separated Epithelium and Stroma From Benign Hyperplastic Prostatic Tissue

Abstract: Using arginase and hydroxyproline as biochemical markers, the yields and homogeneity of separated epithelial and stromal tissues from surgically removed benign hyperplastic prostate glands have been assessed. On the basis of these markers, about 30% of epithelial and 95% of stromal tissues were recovered. Dehydroepiandrosterone sulphate sulphatase activity was found predominantly in the epithelium, whereas testosterone 5alpha-reductase activity was predominantly in the stroma.

“…The radiometrie assay described by Wallace & Grant (1975a) and Cowan, Cowan, Grant & Elder (1977) was used with adaptations to suit the tissues under investiga¬ tion. Weighed tissue was chopped with scissors, dropped into liquid nitrogen and pulverized in a stainless-steel press also cooled in liquid nitrogen.…”

“…To each tube 100 µg 5a-androstane-3a,17ß-diol, a possible further metabolite of DHT, were also added, to act both as a carrier during purifica¬ tion and to assess procedural losses. Steroids were extracted with chloroform, acetylated with pyridine-acetic anhydride (2:1, v/v) and purified by thin-layer chromatography on alumina (Cowan et al, 1977 After the initial measurements, 4 pooled samples of the DHT fractions and 4 pooled samples of the androstanediol fractions were made from each of the 4 tissues. The DHT (as acetate) was recrystallized to constant isotope ratio after carrier DHT acetate had been added.…”

“…There are strong indications that primarily androgens control the function of the stromal part of the human prostate. In 1977, Cowan et al [5]for the first time reported that 5α–reductase activity was found predominantly in the stromal tissue of the prostate. Using a mechanical technique for separating the glandular and stromal parts, Krieg et al [25], Romijn et al [26], and Bruchovsky and Lieskovsky [27]found that the 5α–reductase activity in the stromal fraction of the prostate was considerably higher than in the glandular fraction.…”

“…It was assumed that DHT was only made locally in the normal and ‘hyperplastic’ prostatic epithelial cells by 5α–reductase. Recent studies, however, suggest that intraprostatic DHT is produced exclusively in prostatic stromal and basal cells [5, 6, 7]. Thus, DHT effects on the epithelial cells are likely to be paracrine rather than autocrine in nature.…”

“…It has been reported that differences in growth requirements between epithelial and stromal cells, keratin-positive epi thelial cells and fibroblast-like stromal cells were found in epithelial and stromal tissues, respectively [27], More over, the contents of DNA. RNA and protein in epithelial tissue were significantly lower than that in stromal tissue [28], Since the different constitution of the epithelium and stroma of BPH caused different thermal responses, the epithelium was found to be more thermo-sensitive than the stroma. The conversion of the a helix to a ran dom coil or a P structure in the protein has also been stud ied [29][30][31], This interconversion might have occurred by changing the environmental factors such as dehydration, pH, solvent, temperature, chemicals, and higher pressure [30,31].…”
