Biochemical mechanism underlying hypertriglyceridemia and hepatic steatosis/hepatomegaly induced by acute schisandrin B treatment in mice

Abstract: BackgroundIt has been demonstrated that acute oral administration of schisandrin B (Sch B), an active dibenzocyclooctadiene isolated from Schisandrae Fructus (a commonly used traditional Chinese herb), increased serum and hepatic triglyceride (TG) levels and hepatic mass in mice. The present study aimed to investigate the biochemical mechanism underlying the Sch B-induced hypertriglyceridemia, hepatic steatosis and hepatomegaly.MethodsMale ICR mice were given a single oral dose of Sch B (0.25–2 g/kg). Sch B-in… Show more

“…Although the synthesis of apo B is reduced, it finally leads to the deposition of TG in the liver and the onset of fatty liver (Fan et al., 2018). TF treatment increased serum TG while lowering serum TC levels, which may be caused by the increased serum apolipoprotein B48 induced by TF (Zhang et al., 2017). Nevertheless, TF had significantly decreased hepatic TG and TC levels in mice.…”

“…Hypertriglyceridemia is closely associated with insulin resistance [37]. The liver and adipose tissue are the main sites of triglyceride synthesis [38]. Ang II/AT1R signaling, which is upregulated in T2D, is involved in the suppression of the protein expression and enzymatic activity of hepatic N-deacetylase (NDST) [39] This is a key regulatory enzyme involved in heparan sulphate (HS) biosynthesis in the diabetic state and suppression of hepatic NDST was suggested to lead to diabetic dyslipidemia [40][41][42].…”
