Biodegradable Gene Carriers Containing Rigid Aromatic Linkage with Enhanced DNA Binding and Cell Uptake

Abstract: The linking and modification of low molecular weight cationic polymers (oligomers) has become an attracted strategy to construct non-viral gene carriers with good transfection efficiency and much reduced cytotoxicity. In this study, PEI 600 Da was linked by biodegradable bridges containing rigid aromatic rings. The introduction of aromatic rings enhanced the DNA-binding ability of the target polymers and also improved the stability of the formed polymer/DNA complexes. The biodegradable property and resulted DN… Show more

“…Higher degrees of modification resulted in more zPEI being required to fully bind the DNA. For example, unmodified PEI required a bPEI/DNA ratio of 0.375 (w/w) in good agreement with prior literature . In contrast, to fully condense the DNA, zPEI 3, 6.5, 9, and 11.5 required polymer/DNA ratios of 0.625, 0.625, 0.75, and 1.125 (w:w), respectively.…”

“…Further modification of this polymer with substance P (SP) as the targeting ligand through PEG produced a conjugated polymer that had excellent biocompatibility, ROS cleavability ad transfection efficacy and gene silencing activity in vitro and satisfactory antitumor activity in vivo. A recent study shows that linking of 600 Da PEI with biodegradable bridges of aromatic rings could enhance the DNA binding ability of low MW PEI and increase its stability and transfection efficacy [118].…”

“…Alongside several synthetic methods ( 16 19 ), the administration of the antimalarial drug chloroquine during transfection has been commonly employed to promote intracellular plasmid DNA (pDNA) unpackaging ( 20 ). In addition to promoting endosomal escape ( 21 23 ), chloroquine has been shown to facilitate unpackaging of pDNA by competitive binding ( 24 , 25 ).…”
