Biodegradable polymer with collagen microsponge serves as a new bioengineered cardiovascular prosthesis

Abstract: The poly(lactic-co-glycolic acid)-collagen microsponge patch with and without precellularization showed good histologic findings and durability. This patch shows promise as a bioengineered material for promoting in situ cellularization and the regeneration of autologous tissue in cardiovascular surgery.

“…Mechanistically, a role of the cytoskeleton is to maintain a cell's internal tension, which is altered by changes in cytoskeletal structure and by exposure to external forces (e.g., FSS) (14). Cytoskeletal changes induced by micropatterned lanes or by FSS result in an increase in cellular internal tension and stiffness (15,16). Because the distribution of mechanical forces is critical to eliciting specific mechanotransduction pathways, the internal tension induced by the aligned cytoskeletal structure, with or without FSS, may play an important role in regulating EC functions.…”

“…Alternatively, a biodegradable graft containing collagen microsponge was fabricated and tested, with and without preseeding. 183 In both cases (SIS and collagen microsponge) there was no thrombus formation, the scaffold was absorbed, and there was endothelialization, parallel smooth-muscle cell alignment, elastin, and collagen fibers. These results suggested that the patch promoted in situ cellularization and regeneration of autologous tissue.…”

“…An important biological role is related to HA oligosaccharides that stimulate cytokine secretion and endothelial cell proliferation (60)(61)(62)(63)(64). Moreover, recent studies indicate that HA may inherently stimulate endothelialization, favoring the recruitment of endothelial cells and endothelial progenitor cells (EPCs) (43,45). EPCs can migrate and adhere inside HA-based biomaterials, maintain their preendothelial phenotype, and express angiogenic factors, especially within the first week of growth (63,65).…”
